36H:一種新的酪胺酸酶抑制劑及其機轉

摘要


在本研究中發現36H是一種具有良好的的抗氧化及抑制酪胺酸酶活性特質的化合物。實驗中以DPPH自由基清除能力、金屬離子螯合能力以及鐵離子還原抗氧化力這三種來測定36H的抗氧化能力,發現在自由基清除和鐵離子還原能力上有良好的表現。另外,實驗中也以蘑菇酪胺酸酶,小鼠黑色素瘤以及人類表皮黑色素細胞進行酪胺酸酶活性抑制實驗,發現36H具有明顯的抑制酪胺酸酶活性的能力,而且不會顯著的使細胞存活率下降,此外藉由定量即時聚合酶鏈鎖反應和西方墨點法實驗得知36H可以下調部分黑色素合成相關之蛋白質表現,包含Tyrosinase、Pmel17、Myo5a以及Mreg。這些蛋白質影響黑色素生成、黑色素小體成熟以及黑色素小體的傳輸過程,因此,本實驗結論推斷36H有機會發展成新的皮膚美白試劑。

36H is an active component that shows aniti-oxidantion and anti-tyrosinase activity. It was screened for antioxidant abilities using: 1,1-diphenyl-2-picrylhydrazul (DPPH), metal chelating power test and ferric reducing/antioxidant power (FRAP) examination. The result revealed that 36H had potential anti-oxidative capabilities. In addition, 36H was evaluated for tyrosinase inhibitory activities on in vitro mushroom, B16-F10 mouse melanoma cells and human melanocyte cells. Results showed that 36H owned significant inhibitory effects on the enzyme activities and no obviously decreased cell viability. Besides, through the quantitative real time polymerase chain reaction (QRT-PCR) and western blot, we found it could down regulate melanogenesis, melanosome mature and transport related RNA and protein, Tyrosinase, Pmel17, Myo5a, Mreg and so on. Therefore, 36H could be possibly developed to a new skin whitening agent
