含銀之介孔矽基鈣磷酸鹽生醫玻璃之合成與抗菌分析

Synthesis and antibacterial evaluation of silver-containing mesoporous silica-based calcium phosphates bioglass powders

摘要


本研究使用蒸發-誘導自組裝法 ( evaporation induce self-assembly process, EISA ) 以 80SO2-15CaO-5P2O5-x Ag,x=0, 0.001, 0.01, 0.1, 1, 5, 10 為比例合成含銀之介孔矽基鈣磷酸鹽生醫玻璃 ( silver-containing mesoporous silica-based calcium phosphates, m-SCP-Ag ),並使用牙周病常見之放線桿菌 ( Aggregatibacter actionmycetemcomitans ) 作為實驗菌株,以粉碇擴散實驗 ( disk-diffusion method ) 評估其抗菌性。在 X 光繞射圖譜中發現m-SCPAg1、m-SCPAg5 及 m-SCPAg10 等材料皆具面心立方 ( Face-centered cubic ) 結構之銀結晶。其比表面積分別為 307.6、280.0 及 264.5 m2/g,且隨著銀比例之增加而下降。在浸漬模擬體液 ( simulated body fluid, SBF ) 24、48、72 及 168 小時後可觀察到氫氧基磷灰石 ( Hydroxyapatite ) 結構。m-SCPAg 皆對革蘭氏陽性之金黃色葡萄球菌及革蘭氏陰性之大腸桿菌、放線桿菌具抗菌性,m-SCPAg 10 對金黃色葡萄球菌具最佳之抑菌效果,其抑菌圈尺度為 19.0mm;m-SCPAg1 對大腸桿菌、放線桿菌具最佳之抑菌效果,其抑菌圈尺度分別為 21.0mm 與 16.5mm。含銀之介孔矽基鈣磷酸鹽生醫玻璃具有幫助骨質再礦化與主動抑菌效果,期能成為牙科候選材料,幫助牙周病患者回復流失之骨質,並減緩植牙手術所造成之植體周圍炎的發生。

In this study, the composition ratios of Si, Ca, P, and Ag were 80, 15, 5, and x, where x=0, 0.001, 0.01, 0.1, 1, 5, or 10. 80SO2-15CaO-5P2O5-x Ag power was prepa red by evaporation induced self-assembly ( EISA ) process. Aggregatibacter actionmycetemcomitans ( A. actionmycetemcomitans ) is an important member of the subgingival microbiota in case of periodontitis disease, which was chosen and used to evaluate the antibacterial properties of silver-containing mesoporous silica-based calcium phosphates bioglass ( m-SCP-Ag ) powders by disk-diffusion method. The m-SCPAg1, m-SCPAg5, and m-SCPAg10 samples all have face-centered cubic silver. The surface areas for the m-SCP, m-SCPAg1, m-SCPAg5, and m-SCPAg10 samples were 348.9, 307.6, 280.0, and 264.5 m2/g, respectively. Clearly, the surface area decreased with an increasing Ag ratio. On the other hand, samples after being immersed in simulated body fluid ( SBF ) for 24, 48, 72, and 168h, the hydroxylapatite structure was observed. The microbiological assay revealed that the all m-SCPAg samples had an antibacterial effect against E. coli, S. aureus, and A. actinomycetemcomitans. m-SCPAg10 showed the largest diameter ( 19.0mm ) of the inhibition zone for S. aureu, and m-SCPAg1 showed the largest diameter ( 21.0mm, and 16.5 mm ) of the inhibition zone for E. coli, and A. actinomycetemcomitans. Silver-containing mesoporous silica-based calcium phosphates bioglass with bone regeneration and anti-bacteria ability, would release the illness and prevent the peri-implantitis.
