熱與紫外線A之交互作用對於人類真皮纖維母細胞的影響:著重皮膚光老化的研究

The effect of interaction of heat and UVA on human dermal fibroblasts: implications on skin photoaging.

摘要


太陽光依照其波長可分為可見光、紅外線和紫外線 (ultraviolet; UV)。紫外線又分為紫外線A (UVA)、紫外線B (UVB) 和紫外線C (UVC)。UVA可以穿透表皮層到達真皮層,真皮層主要是由纖維母細胞和彈性纖維所組成。此外,纖維母細胞是皮膚老化的關鍵細胞。眾所皆知,太陽光中的UV是皮膚光老化主要的原因,過去的研究顯示,UVA照射所誘導產生的ROS能夠增加MMP-1的產生並增加膠原蛋白的分解,最終導致皮膚的老化。最近,全球暖化在全世界變成非常重要的環境議題,環境溫度的增加對人類健康上的影響逐漸吸引了許多的關注。人類日常生活中暴露於環境熱是不可避免的,然而,人們卻忽略這個重要的問題。先前的研究顯示,兩種不同形式的能量也就是UV和熱可經由不同的途徑導致皮膚老化。然而,在皮膚光老化中UV和熱的相互作用對於皮膚光老化的影響卻很少被討論。因此在本研究中,我們擬使用培養之人類真皮纖維母細胞human dermal fibroblasts (HDFs) 作為研究目標進行光老化之探討。實驗組別將包含控制組、單獨39°C熱處理、單獨3 J/cm2 UVA輻射、3 J/cm2 UVA照射後39°C熱處理 (UVA-Heat) 和 39°C熱處理後3 J/cm2 UVA照射 (Heat-UVA) 等五組。研究結果顯示,與Heat-UVA組比較,UVA-Heat組有較低的type I collagen 和較高matrix metalloproteinase-1 (MMP-1) 的表現。此外,與Heat-UVA實驗組的比較之下,UVA-Heat 組會誘導產生較高的reactive oxygen species (ROS) 的產生。我們的研究結果顯示,除了UVA的因素之外,在發展減少皮膚光老化的保護策略中,環境熱也應該被考慮進去。

Sunlight can be divided into visible light, infrared light and ultraviolet light (UV). UV irradiation can be divided into UVA, UVB, and UVC. UVA irradiation can penetrate into the dermis layer. The dermis layer is mostly composed of fibroblasts and elastic fibers. In addition, dermis fibroblasts are the key cellular component of skin aging. It is known that UV in sunlight is the major cause of skin photoaging. Previous studies revealed that the generation of ROS after UVA irradiation increased MMP-1 production and subsequently lead to increased collagen breakdown, and eventually resulted in premature skin aging. Recently, global warming has become a very important environmental agenda all over the world. The impacts of increased environmental heat on human health have attracted much attention. Exposure to environmental heat is inevitable in the daily life of human beings. However, people often ignore this important point. Previous studies have shown that two different forms of energy namely UV light and heat, can lead to skin aging through different pathways. However, the effects of the interaction of UV light and heat on skin photoaging is rarely discussed. Therefore, in this study, we set out to explore the effects of the interaction of UVA and heat on skin aging by using human dermal fibroblasts (HDFs) as study target. The experimental groups included control group, 39°C heat treatment only, 3 J/cm2 UVA irradiation only, 3 J/cm2 UVA irradiation followed by 39°C heat treatment (UVA-Heat), and 39°C heat treatment followed by 3 J/cm2 UVA irradiation (Heat-UVA). The results demonstrated that UVA-Heat treated HDFs showed significantly lower type I collagen and higher matrix metalloproteinase-1 (MMP-1) expressions as compared to Heat-UVA treated group. In addition, UVA-Heat treatment induced higher reactive oxygen species (ROS) production as compared to Heat-UVA treated group. The results suggested that in addition to UVA irradiation, environmental heat should also be taken into consideration while developing protective strategy on reducing skin photoaging.
