土楠(Cryptocarya concinna Hance syn.: Cryptocarya konishii Hayata ex Kawakami),為樟科(Lauraceae)之中型常綠喬木,分布於華南和台灣。我們研究室對台灣產植物進行抗結核活性篩選,發現土楠莖部具有抗結核活性。由土楠莖部乙酸乙酯可溶部已分離出五個新化合物,包括一個chalcone: concicryptol (1);一個isocoumarin: concinol (2);三個flavanones: 混合物 cryptoflavanonic acid?nA (3)和cryptoflavanonic acid?nB (4), 5- dehyrocryptoflavanone A (5),以及十七個已知化合物:包括兩個chalcones: cryptocaryone (18)、infectocaryone (95);三個isocoumarins: , 5-chloro-8-hydroxy-6-methoxy-3-methylisocoumarin (6)、8-hydroxy-6,7- dimethoxy-3-methylisocoumarin (10)、5-methylmellein (15);七個flavanones: cryptoflavanones C?n?v8?w、cryptoflavanones D?n?v9?w、cryptoflavanone A (11)、cryptoflavanone B (12)、pinostrobin (14)、cryptocaryanone A (97)及cryptochinone B (163);三個steroids: 混合物??-sitosterol (77)和stigmasterol (175), ??-sitostenone (174);一個benzenoid: methyl cinnamate (13)以及一個fatty acid: stearic acid (16)。上述化合物之結構,經由各種圖譜分析及文獻數據比對予以確認。 將所分離到的純化合物進行體外抗結核菌活性測試,其中化合物1, 18, 95, 97及163具有抗結核菌活性,其MIC 值分別為10.0、10.5、12.0、8.0及15.0 mg/mL。
Cryptocarya concinna Hance (syn.: Cryptocarya konishii Hayata ex Kawakami; Lauraceae) are medium-sized evergreen trees, which distributed in south China and Taiwan. In our studies on the antitubercular constituents of Formosan plants, many species have been screened for in vitro antitubercular activity, and C. concinna has been found to be one of the active species. Investigation of the EtOAc-soluble fraction of the stems of C. concinna has led to the isolation of five new compounds, including one chalcone, concicryptol (1), one isocoumarin, concinol (2), and three flavanones, mixtures of cryptoflavanonic acids?nA (3) and?nB (4), 5-dehyrocryptoflavanone A (5), along with 17 known compounds, comprising of two chalcones: cryptocaryone (18) and infectocaryone (95), three isocoumarins: 5-chloro-8-hydroxy-6-methoxy-3- methylisocoumarin (6), 8-hydroxy-6,7-dimethoxy-3-methylisocoumarin (10)、5-methylmellein (15), seven flavanones, cryptoflavanones C?n?v8?w, cryptoflavanones D?n?v9?w, cryptoflavanone A (11), cryptoflavanone B (12), pinostrobin (14), cryptocaryanone A (97) and cryptochinone B (163), three steroids: mixtures of ??-sitosterol (77) and stigmasterol (175), ??-sitostenone (174), one benzenoid: methyl cinnamate (13) and one fatty acid: stearic acid (16). The structures of these isolates were determined through spectral analyses and comparison of their physical and spectral data with literatures. In the in vitro antitubercular activity assay of these isolates, compounds 1, 18, 95, 97 and 163 exhibited antitubercular activities with their MIC values of 10.0、10.5、12.0、8.0 and 15.0 mg/mL, respectively.
