對於有症狀開放性動脈導管症的新生兒,接受Indomethacin水溶液或啤酒溶液,在血清Indomethacin的濃度測量可藉由快速、準確的高效能液相層析法來做分析。利用50% 乙腈溶液從血清中萃取出Indomethacin和內部標準品2,4,5-Trichloronitrobenzene。 所使用的層析柱為 Lichro CART Rp 18e管柱 (55 × 4 mm ; 3µm),移動相是溶液A [water -acetonitrile- acetic acid (44:950:6)]和溶液B [water-acetonitrile-acetic acid(950:44:6)],以55:45混合。層析柱流出物是在波長254 nm被監測。而Indomethacin可被監測的最低濃度為0.2 µg/ml。具有開放性動脈導管症的新生兒給予Indomethacin (0.2mg/kg/dose) 來治療,每12小時間隔給予,共3個劑量。本實驗採隨機選擇,無控制雙盲實驗,給予病人Indomethacin的水溶液或啤酒溶液,在給予劑量後的8小時抽取病人血液做Indomethacin的血清濃度測定。
A rapid, accurate high-performance liquid-chromatographic procedure is intended for measuring indomethacin in serum from neonates who are receiving orally water solution or beer solution of indomethacin for symptomatic patent ductus arteriosus. Indomethacin and an internal standard (2, 4, 5-trichloronitrobenzene) are extracted from serum with acetonitrile. A LiChro CART Rp 18e column ( 55 × 4 mm ; 3mm) and a mobile phase of solution A [water-acetonitrile-acetic acid (44:950:6)] and solution B [water-acetonitrile-acetic acid (950:44:6)] in a ratio of solution A and solution B ( v/v =55:45) were used. The column effluent is monitored at 254 nm. The indomethacin can be detected in concentration as low as 0.2 mg/ml. Preterm infants with patent ductus arteriosus were given indomethacin (0.2 mg/kg/dose) at 12 h intervals up to three times, either orally water solution or beer solution of indomethacin, in an uncontrolled, double-blind, randomized study. Serum indomethacin concentrations were determined in blood samples 8 h later.
