液相層析法對人體血漿中Coenzyme Q10之分析及應用

摘要


本論文以液相層析法對人體血漿中coenzyme Q10之含量分析,探討正常人及鬱血性心衰竭病人之coenzyme Q10含量分析,並對26位冠狀動脈疾病合併有高血脂病人,服用atorvastatin降血脂,探討服藥前、服藥後3個月、停藥後第2天及停藥後第3天,病人體內血漿中coenzyme Q10之含量分析。本研究以1-butanol為萃取溶媒,以甲醇:正己烷=85:15為移動相溶媒,coenzyme Q9為內部標準,以逆相C18層析管柱分離,以紫外光檢測器275 nm為檢測波長。正常人其體內coenzyme Q10濃度為0.7-1.0 ug/mL,對於鬱血性心衰竭之病人其血漿中coenzyme Q10含量比正常人低,而冠狀動脈疾病合併有高血脂病人,服用atorvastatin 3個月後,病人血漿中coenzyme Q10之含量大約降低了60%,服藥前及服藥後coenzyme Q10濃度,有明顯差異(p< 0.001),因此鬱血性心衰竭之病人、冠狀動脈疾病合併有高血脂而服用atorvastatin之病人,其體內coenzyme Q10含量比正常人低,所以建議補充coenzyme Q10,以提高體內coenzyme Q10之濃度。 本法可測定血漿中coenzyme Q10之線性範圍是0.1-4 ug/mL,其萃取相對回收率大於93.33%,證明本法具有良好之定量可靠性,確可應用於人體血漿中coenzyme Q10之含量測定。

Plasma coenzyme Q10 levels in normal, congestive heart failure (CHF) patients and coronary artery disease combined hyperlipidemia (CAD-HL) patients were determined using high performance liquid chromatography. The coenzyme Q10 concentration of 26 CAD-HL patients were determinaed including pre-dosing atorvastatin, after-dosing atorvastin, 2 days after withdraw atorvastin and 3 days after withdraw atorvastin. Coenzyme Q10 in human plasma was extracted with 1-butanol. The extracted was separated on a LichroCART?? RP-18 column (55 × 4 mm, 3 um) with methanol-hexane (85:15, v/v) as mobile phase. The column eluate was monitored at UV detector 275 nm. The concentration of coenzyme Q10 in CHF patients were lower than normal (07-1.0 ug/mL). The concentration of coenzyme Q10 in CAD-HL patients were decreased about 60% after-dosing atorvastatin 3 months, so pre-dosing vs after-dosing atorvastatin has significant differences (p<0.001). For CHF patients and CAD-HL patients take atorvastatin, the concentrations of plasma coenzyme Q10 were decreasing and lower than normal. Therefore, we suggested them to take coenzyme Q10 for increasing the coenzyme Q10 concentrations. From the study the levels of coenzyme Q10 in human plasma was 0.1-4 ?慊/mL. The relative recovery was over 93.33%. Application of the method to the analysis of coenzyme Q10 in human plasma proved to be feasible.
