本研究討論當歸萃取物對促進fenofibrate之皮膚穿透性的效果。首先,製備含當歸萃取物2、4、6、8 % (w/v)之7.5 % (w/v) fenofibrate懸浮劑,進行體外皮膚穿透試驗,測定透過皮膚之fenofibrate的累積量,結果顯示所有處方約同程度地、有效地增強 (p<0.05) fenofibrate之皮膚穿透性。 其次,製備含當歸萃取物2、4、6、8 % (w/v)之7.5 % (w/v) fenofibrate凝膠劑,進行體外皮膚穿透試驗,測定透過皮膚之fenofibrate的累積量,結果顯示當歸萃取物之含量為2 %時,無法有效提昇fenofibrate之皮膚穿透性,但當歸萃取物之含量增為4、6及8 % 時,則fenofibrate之皮膚穿透性有意義地增加 (p<0.05),且呈現劑量依存性 (dose-dependent)。 所有處方之時間與累積皮膚透過量之間呈現零階次動力學的關關係,以Fick’s law of diffusion 處理之後,發現當歸萃取物之添加,有意義地增強fenofibrate的穿皮速度,且縮短滯留時間 (lag time)。 從本研究之結果,可看出當歸萃取物有意義地增加fenofibrate的皮膚穿透性,但作用開始發揮時間慢。這些結果也顯示當歸萃取物可作為經皮吸收促進劑使用,但宜添加足以改善作用開始發揮時間的第二種經皮吸收促進劑。
In this study, we evaluated the effectiveness of the fenofibrate skin penetration with Tooki extract. Firstly, we prepared 7.5 %(w/v) fenofibrate suspension containing 2、4、6、8 % (w/v) of Tooki extract, and the skin penetration experiments were carried out, the cumulative mount of fenofibrate penetrating through skin into receptor side was examined. The results shown all of formulations significantly (p<0.05) enhanced the skin penetration of fenofibrate with the same degree. Secondly, we prepared 7.5 %(w/v) fenofibrate gel containing 2、4、6、8 % (w/v) of Tooki extract, and the skin penetration experiments were carried out, the cumulative mount of fenofibrate penetrating through skin into receptor side was examined. The results shown the system with 2 % of Tooki extract did not significantly (p>0.05) enhanced the skin penetration of fenofibrate, but the 4、6、8 % (w/v) of Tooki extract systems significantly (p<0.05) enhanced the skin penetration of fenofibrate. According to the penetration profiles of all formulations, it was shown that the skin penetration of fenofibrate followed the zero-order kinetics with Tooki extract. Therefore, the data can be treated according to the Fick’s law of diffusion. Addition of Tooki extract significantly enhanced the penetration rate. The lag time were also reduced. Form the results in this study, the Tooki extract significantly increased the fenofibrate skin penetration, but the onset was slow. Furthermore, these results demonstrated that the Tooki extract was suitable for use as a skin penetration enhancer, but it was necessary to incorporate a second skin penetration enhancer which will improve the onset time of the system containing fenofibrate。
