單元不飽和脂肪酸對調節性T細胞表現的影響

摘要


飲食油脂對於免疫調節之研究,大多著重於多元不飽和脂肪酸,但少有研究探討單元不飽和脂肪酸;本實驗室之前研究計畫結果顯示,飲食單元不飽和脂肪酸可增加調節性T細胞激素的分泌,已知調節性T細胞,在治療過敏性氣喘方面,扮演重要的角色。因此,本研究分為體外及體內實驗二個部分,第一部分以體外實驗探討單元不飽和脂肪酸對Treg的影響,實驗採用亞麻油酸及油酸為實驗材料,處理脾臟及腸繫模淋巴結細胞。結果顯示,單獨油酸處理,會促使脾臟細胞增生的效用。在ConA 刺激下,脂肪酸會抑制淋巴細胞增生,而油酸之抑制效應低於亞麻油酸;但是油酸處理下,CD4+CD25+及CD4+CD25+FOXP3+的比例低於同劑量之亞麻油酸組,且增加CD4+CD25-比例,並在低劑量油酸處理時,增加TGF-β分泌量。 第二部分為探討單元不飽和脂肪酸對過敏性氣喘小鼠體內Treg的影響,本實驗以OVA腹腔注射與吸入性致敏,誘發過敏性氣喘小鼠研究模式,並給予含有5%、20%的大豆油或橄欖油粉狀飼料,八周。結果顯示,20%橄欖油可以顯著減緩呼吸道過度反應(AHR)及降低CD4+CD25+FOXP3+的比例、降低TGF-β和IL-10分泌量。綜合本研究之結果得知,飲食橄欖油可以減緩過敏性氣喘呼吸道過度反應、降低調節性T細胞的比例及其細胞激素。而主要功效來自於橄欖油中之油酸。

The study of immunomodulatory effects of dietary lipid were investigated the diet rich in polyunsaturated fatty acid (PUFA) but less attention has been paid to the effects of monounsaturated fatty acid (MUFA). In our pervious study, the MUFA-rich diet increased the regulatory T (Treg)-cells cytokine production by Con A-stimulated splenocytes from BALB/c mice. It has been know regulatory T-cells play an important role in treatment allergic asthma. Therefore, we like to study the effects of MUFA on Treg expression in vitro first. Oleic acid enhanced the Treg expression in splenocytes and TGF-β secretion. And to investigated the effects of MUFA-rich diet Treg expression in vivo. BALB/c mice sensitized intra-peritoneally and challenged with OVA were treated with different percent soybean oil or olive oil for 8 weeks. And CD4+CD25+FOXP3+ percentage, significantly. And decreased regulatory cytokines. Overall these data suggest that olive oil -rich diet could lower airway hyper-responsiveness and Treg expression in allergic asthma. Olive oil could be a health dietary oil for allergic asthma.
