Analysis of Organic Solvent Mixtures in Urine Using Hollow Fiber Headspace Liquid-phase Microextraction and Gas Chromatography with Flame Ionization Detector

摘要


接著劑工廠使用如苯、甲苯、二甲苯、正己烷、正辛烷、正庚烷、甲基環己烷多種有機溶劑,勞工在作業場所暴露這些不同種類的溶劑,這些物質會被轉變成不同的代謝物,因此勞工暴露有機溶劑的生物偵測是評估化學溶劑毒性影響的重要步驟。 本研究所使用的中空纖維頂空液相微萃取(HF-HS-LPME)結合氣相層析儀/火焰離子偵測器(GC-FID)的方法來分析尿液中多種有機溶劑,萃取的參數如萃取溶劑、攪拌速率、鹽類添加、萃取時間、萃取溫度及pH值等對萃取效率有重要的影響。最佳萃取條件為吸附萃取溶液:2.0 ?愮正辛醇、萃取時間10分鐘、萃取溫度40℃、pH 7.0及不添加NaCl。 用此方法已成功分析在接著劑工廠暴露勞工所收集的尿液樣本,結果顯示下班前後的勞工尿液中有機溶劑的含量比上班前的勞工尿液中的有機溶劑的含量高,此最佳的萃取方法可適用於尿液中多種有機溶劑的分析。

Multiple organic solvents, such as benzene, toluene, xylene, n-hexane, n-heptane, n-octane and methylcyclohexane, were used in an adhesive of factory. Workers exposure to these different kinds of solvents in the workplace. These substances will be translated into different kinds of metabolites. Therefore, biological monitoring of workers exposed to organic solvents is important step toward the evaluation of the toxicological effect of chemical solvents. In this study, a hollow fiber headspace liquid phase microextraction(HF-HS-LPME) and gas chromatography-flame ionization detector(GC-FID) method for the analysis of multiple organic solvent mixtures in urine was described. Extraction parameters such as extraction solvent selection, stirring rate, salt addition, extraction time, extraction temperature and pH were found to have significant influence on extraction efficiency. The optimized conditions were 2.0 ?愮 1-octanol extraction solvent, 10 min extraction time, 40℃, 500 rpm stirring rate, pH 7.0 and without NaCl addition. Urine samples were obtained from exposed workers in the adhesive factory and were successfully analyzed using the proposed method. The results showed that the amounts of organic solvent mixtures in the end of shift urine samples were higher than those of in the pre-shift urine samples. This optimized method is suitable for the analysis of multiple organic solvent mixtures in urine samples.
