宿主膜系重組蛋白,PSTPIP2, 調控丙型肝炎病毒的複製與其誘導的膜網形成

摘要

丙型肝炎病毒（HCV）感染後會重組細胞內膜以形成新的胞器供病毒複製; 然而目前對於病毒與細胞蛋白如何交互作用而造成細胞內膜重組的機制的了解仍然相當有限。為了更進一步探究有哪些人類蛋白參與丙型肝炎病毒的複製, 我們將shRNA使用慢病毒 (lentivirus) 送入細胞的技術篩選出與丙型肝炎病毒複製相關的蛋白; 經過一連串的實驗確認後, 我們發現具有使細胞膜變形能力的人類蛋白PSTPIP2 參與丙型肝炎病毒的複製。 我們進而發現抑制PSTPIP2的表現會使丙型肝炎病毒在細胞內的蛋白和核醣核酸表現降低, PSTPIP2的抑制也會減少丙型肝炎病毒和病毒蛋白NS4B誘導的網膜形成。

我們更發現PSTPIP2因突變而缺乏細胞膜變形能力的蛋白也同時失去了促進病毒核醣核酸與蛋白複製的能力, 這個發現更加確認PSTPIP2使細胞膜變形的特性會影響丙型肝炎病毒的複製。因此, 我們認為PSTPIP2 不但在病毒誘導的網膜形成扮演關鍵的角色, 進而還會影響丙型肝炎病毒的複製。
