應用核磁共振技術確認梳狀高分子奈米微胞材料之合成

摘要


核磁共振波譜儀 (NMR) 是一種可用於分析有機分子及高分子微觀化學結構的方法,不論是固態、液態、膠態都可分析。依脈衝程序的不同分為一維、二維、多維圖譜,本篇研究的分析物是屬液態,運用特定功能的脈衝程序(例:一維圖譜,二維同核、異核圖譜),透過化學鍵傳遞的J-偶合作用或空間偶極作用來推測分子內各原子核間的鍵結情形與幾何結構。 高分子微胞 (Polymeric Micelle) 藥物傳遞系統為一具有核一殼型(Core-shell) 型態的藥物載具用於高分子藥物傳遞之為一具兩性 (Amphiphilic)的分子鏈段所組成,其中一端帶有親水鏈段 (Hydrophilc chain),另一端則帶有親油鏈段 (Hydrophobic chain) ,當兩性高分子於水溶液中的濃度超過臨界微胞濃度 (CMC) 時,分子中的疏水鏈段會透過凡得瓦爾力 (van der Waal’s force) 相互作用,經由自組裝形成高分子奈米微胞結構,使分子形成疏水基在內而親水基在外之共聚物,若將藥物置於奈米微胞中則可作為藥物載體 (Drug Carrier) 以控制藥物緩解的方式與時間。 利用兩性高分子材料作為合成藥物載體之原料,在此實驗中為使用高解析度的液態核磁共振儀來分析 HA-g-PCL 與 mPEG-g-PCL 之合成機制。本篇研究中我們運用氫譜、碳譜、 Diffusion、 COSY、 editHSQC、 HMBC、 1D-NOE等實驗技術來鑑定原物料純度是否適用,並在中間體及合成產物上做合成結構上的確認。

Nuclear magnetic resonance spectroscopy (NMR) is a method for analyzing both small molecules and polymer chemical structure. It’s application includes among solid, liquid, and gel states samples. The pulse sequences may be classified as one-dimensional, two-dimensional, multi-dimensional . The samples for this study belongs to liquid state, and we used the function specific pulse sequence for this study. Pass through the J-coupling effect of chemical bond or the space dipolar interaction to speculate the bond status and geometric structure of the atomic nucleus in intramolecular. Polymeric micelle for drug delivery system is a nanoparticles ball with core-shell structure. The polymeric micelle is composed of amphiphilic blocks. One of which is hydrophilic chain and the other is hydrophobic. When the concentration of amphiphilic polymer in aqueous solution is grater than the critical micelle concentration (CMC), the hydrophobic chain parts pass through the interaction of van der Waals force. By the way, the self-assembly structure of polymer nano micelles forms the copolymer of including inner hydrophobic core and outer shell hydrophilic. And then the nano micelles as drug carriers to control the manner and time of drug ease. Use two kinds of hydrophilic polymers as the raw materials of synthetic drug carriers. In this experiment, use high-resolution liquid NMR to analyze the synthesis mechanism of HA-g-PCL and mPEG-g-PCL. First of all the used of hydrogen spectrum, carbon spectrum, Diffusion, COSY, editHSQC, HMBC, 1D-NOE and other experimental techniques to identify the purity of raw materials were suitable or not and confirmed the synthesized structure of intermediates and products.
