摘要


本論文旨在合成神經鞘胺醇衍生物xyl-14、脂肪酸衍生物Rib-7及唾液酸醣苷神經鞘醯胺質SJG-1之類似物SJG-1A、SJG-2A、SJG-3A、SJG-4A、SJG-5A、SJG-6A。 我們以D-核醣(D-ribose)作為起始物經保護,經Wittig reaction、氧化等七個反應得到脂肪酸衍生物Rib-7。 我們以D-木醣(D-xylose)作為起始物,經由保護、Wittig reaction、SN2取代反應、Staudinger reaction、烯烴複分解反應(olefin metathesis)等十六個反應製備而得神經鞘胺醇衍生物xyl-14。 本論文以化學選擇性醣苷化反應及[1+1+2]的合成策略來合成SJG-1之類似物,目前已完成SJG-1之類似物主架構之合成。

This paper aims to complete the synthesis of sphingosine derivative xyl-14, fatty acid derivative Rib-7 and Ganglioside SJG-1’s Analogues with Various diihydroceramides. We use D-ribose as a starting material to obtain fatty acid derivative Rib-7, seven-step reaction via protection, Wittig reaction and oxidation. We use D-xylose as a starting material to obtain sphingosine derivative xyl-14, sixteen-step reaction via protection, Wittig reaction, SN2 reaction, Staudinger reaction and Olefin metathesis. We used chemoselective glycosylation and [1+1+2] stragety to construct the target SJG-1’s protected analogues.
