台灣香檬精油之皮膚保健功效評估

Evaluating the effects of Citrus depressa essential oil on skin health

摘要


皮膚是人體最大的器官,作為身體和外界環境的屏障,可抵禦外來環境物質的第一道防線,故皮膚保健是相當重要的課題。已有研究指出植物精油具抗氧化和抑菌等生物活性,故本研究即針對台灣香檬 (Citrus depressa Hayata, 縮寫為 CD) 果皮進行精油 (essential oil, 縮寫為 EO) 萃取,並進一步評估其對皮膚之相關保健功效。經 GC-MS 分析 CD-EO 組成分,發現含量最高的三個主要化合物為 (R)-(+)-limonene (38.97%)、γ-terpinene (24.39%) 和 linalool (6.22%)。在 DPPH 和 ABTS 清除自由基測試中,CD-EO 的 Trolox 等價抗氧化能力分析 (Trolox equivalent antioxidant capacity assay, TEAC) 分別為 0.28 和 2.24 mg TE/g。於促癌劑 TPA 誘導小鼠表皮 JB6 P+ 細胞癌化轉型之試驗中,發現精油 (100 μg/mL) 可顯著抑制細胞非貼覆性生長 (p < 0.05),且提高了小鼠表皮 JB6 P+ 細胞內Nrf2及其下游抗氧化酵素 (如 NQO1、HO-1 和 UGT1A 等) 之蛋白質表達量。此外,CD-EO 也對白色念珠菌 (Candida albicans)、綠膿桿菌 (Pseudomonas aeruginosa)、大腸桿菌 (Escherichia coli) 和金黃色葡萄球菌 (Staphylococcus aureus) 等微生物皆有抑制生長之作用。故綜合以上結果,顯示CD-EO具有皮膚保健之潛力,包括抗氧化、化學預防皮膚癌化生成,以及抑制致病微生物生長等作用。

Human skin is the body’s largest organ, and is the first line of defense against the external environment. Hence, maintaining skin healthy is a very important issue. Numerous studies have shown that plant essential oils have certain biological functions, such as antioxidant and antibacterial activities. In this study, we obtained the essential oil from Citrus depressa peel (CD-EO) in Taiwan and further assessed its effects on skin health. The composition of CD-EO was analyzed by GC-MS, and the principal compounds were (R)-(+)-limonene (38.97%), γ-terpinene (24.39%) and linalool (6.22%). In the DPPH and ABTS free radical scavenging assays, the Trolox equivalent (TE) antioxidant capacity assay (TEAC) of CD-EO was 0.28 and 2.24 mg TE/g, respectively, suggesting that CD-EO has antioxidant activity. We also found that CD-EO (100 μg/mL) can significantly inhibited 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced carcinogenic neoplastic transformation of mouse skin JB6 P+ cells (p<0.05). Additionally, CD-EO suppressed the growth of pathogenic Candida albicans, Pseudomonas aeruginosa, Escherichia coli, and Staphylococcus aureus. These results suggested that CD-EO is beneficial for skin health, including antioxidation, skin cancer chemoprevention, and pathogenic microorganisms inhibition.
