台灣香檬萃取物對高脂飼料誘導小鼠 非酒精性脂肪肝之保護效果

摘要


臺灣香檬 (Citrus depressa Hayata, CD) 又名扁實檸檬,是台灣特有的柑橘類水果,為芸香科 (rutaceae) 柑橘屬植物。台灣香檬果皮中含有豐富的多甲氧基黃酮類 (polymethoxyflavones, PMFs),其中又以Nobiletin、Tangeretin 的含量最為豐富,目前已有研究顯示出具有抗發炎、抗癌、抗動脈粥狀硬化以及減少脂肪的生成等功效。本研究乃探討台灣香檬果皮與其活性成分對高脂飲食所誘導之 C57BL/6 小鼠非酒精性脂肪肝之保護功效與作用機制。台灣香檬果皮經烘乾後置備成乙醇萃取物 (ethanolic extract,CD-EE),並將組別分成控制組、高脂飲食組、台灣香檬果皮萃取物低劑量CD-EE (250 mg/ kg bw )以及高劑量組 CD-EE (500 mg/ kg bw )、Nobiletin (50 mg/ kg bw) 組和Tangeretin (50 mg/ kg bw) 組,分別管餵 C57BL/6 小鼠18週後,於第18週犧牲時檢測小鼠心臟血液中alanine aminotransferase (ALT)、aspartate aminotransferase (AST)、三酸甘油脂 (TG)、膽固醇 (TC),並檢測肝臟內 TG和 TC 的含量,CD-EE低劑量組和高劑量組、Tan 組 和 Nob組能夠減少因脂肪變性所導致之肝損傷,使血液中 AST 和 ALT 的活性顯著下降,並且有效的降低肝臟中累積的脂質油滴的大小,讓肝細胞不會受到損傷,且CDEE低劑量組和高劑量組、Tan 組 和 Nob組能夠改善高脂飲食所誘導之肥胖以及緩解階段性的體重增加,並且能夠抑制脂肪組織的生成依照上述實驗成果能作為保健食品開發之依據,以增進台灣特有種柑橘類水果台灣香檬的農作經濟價值。

Taiwan lemon (Citrus depressa Hayata, CD), also known as flat lemon, is a unique citrus fruit in Taiwan, belonging to the citrus genus of Rutaceae (rutaceae). Taiwan bergamot peel is rich in polymethoxyflavones (polymethoxyflavones, PMFs), among which Nobiletin and Tangeretin are the most abundant. Current studies have shown that they have anti-inflammatory, anti-cancer, anti-atherosclerosis and Reduce fat production and other effects. This study was to investigate the protective effect and mechanism of Taiwan lemon peel and its active ingredients on high-fat diet-induced non-alcoholic fatty liver in C57BL/6 mice. Taiwan bergamot peel was prepared into ethanol extract (CDEE) after drying, and the groups were divided into control group, high-fat diet group, and Taiwan bergamot peel extract low-dose CDEE (250 mg/ kg bw ) and high-dose CDEE (500 mg/ kg bw ), Nobiletin (50 mg/ kg bw ) and Tangeretin (50 mg/ kg bw ) groups were administered to C57BL/6 mice for 18 weeks, Detect alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), cholesterol (TC) in the heart blood of mice at the sacrifice of the 18th week, and detect the contents of TG and TC in the liver, CD-EE Low-dose group and high-dose group, Tan group and Nob group can reduce the liver damage caused by fatty degeneration, significantly reduce the activity of AST and ALT in the blood, and effectively reduce the size of lipid oil droplets accumulated in the liver, so that Liver cells will not be damaged, and CDEE low-dose group and high-dose group, Tan group and Nob group can improve the obesity induced by high-fat diet and alleviate the phased weight gain, and can inhibit the formation of adipose tissue According to the above experimental results It can be used as a basis for the development of health food to enhance the agricultural economic value of Taiwan lemon, a unique citrus fruit in Taiwan.
