台灣細本山葡萄抗凝血活性成分之研究

Isolation and Characterization of Anticoagulants from Vitis thumbergii var. taiwaniana.

摘要


台灣細本山葡萄(Vitis thunbergii var.taiwaniana)是台灣民間常用的食材及藥用植物,已有報導顯示具有抗氧化、抗發炎、抗癌及抗凝血的功效。凝血機制依照所牽涉之凝血因子可分為外源性、內源性,可藉由檢測血漿凝血酶原時間prothrombin time (PT)與活化部分凝血活酶時間activated partial thromboplastin time (aPTT)加以區別。此外血小板活性也於凝血機制中扮演重要角色。本研究萃取台灣細本山葡萄內的化合物分析並探討其抗凝血的機制。先將植物根部以80%甲醇萃取,接續以異丙醇鹽析得到異丙醇層主要的成分。藉由矽膠管柱層析,以不同比例之二氯甲烷、甲醇、水進行沖提。得到的萃取物在進行濃縮後,測試其抗凝血活性。具抗凝血活性的萃取物利用液相層析儀進一步分離純化並鑑定主要成分結構。經由液相層析質譜(LC-MS)與核磁共振(NMR)鑑定並與文獻資料比對後,確定抗凝血活性萃取物中之主要成份hopeaphenol、 (+)-viniferol D、(+)-catechin。將分離出之成分進行抗凝血活性分析以確定抗凝血機制。血漿凝血酶原時間(PT)與活化部分凝血活酶時間(aPTT),顯示化合物hopeaphenol、 (+)-viniferol D作用於外源性凝血路徑,再由COX-2和VKOR活性測試得知化合物hopeaphenol、 (+)-viniferol D抑制 VKOR而達到抗凝血的效果。

Vitis thunbergii var.taiwaniana is commonly used in Taiwan's food and medicinal plants, has been reported to have anti-oxidation, anti-inflammatory, anti-cancer and anti-coagulation effect. The coagulation mechanism can be divided into extrinsic, intrinsic, detectable prothrombin time (PT) and activated partial thromboplastin time (aPTT) in terms of the coagulation factor involved. In addition, platelet activity also plays an important role in the coagulation mechanism. In this study, the compounds in the Vitis thunbergii var.taiwaniana were extracted and the mechanism of anticoagulation was explored. The root of the plant was first extracted with 80% methanol, followed by the isopropanol precipitation step to obtain the main components of the isopropanol layer. Separation by silica gel column chromatography. The resulting extract was tested for its anticoagulant activity after concentration. The extract with anticoagulant activity was was using a liquid chromatography to isolated and purified. By LC-MS and NMR identified,the main components of the extract with anticoagulant activity were hopephenol ,(+)-viniferol D and (+)-catechin. By COX-2 and VKOR activity that two compounds hopephenol ,(+)-viniferol D are VKOR inhibition.
