大腸直腸癌生物感測器之研發

Development of a Biosensor for Colorectal Cancer Detection

摘要


大腸直腸癌於西元1981~2010年間,由癌症死亡原因第五位升至第三位,於2010年為主要癌症死因第三位,即使全世界,大腸直腸癌也高居第四位。本研究目的為研發一種短時間內能即時測量大腸直腸癌標的物(CEACAM6)之阻抗變化生物感測器,與其他傳統檢測方法造成的不便、花費時間過長與成本花費過高比較起來,更能達到及時量測、低成本與便利等優點,實現及早發現及早治療之目標。 利用網版印刷技術,將銀膠線路及碳膠保護層印刷於感測器基材上,最後將CEACAM6抗體固定於感測器上製作出CEACAM6生物感測器。經量測後發現生物感測器在610.4Hz~51.76kHz表現出良好的線性度R^2 >0.9,在此頻率範圍內於37.35kHz實呈現最佳值R^2 = 0.9978。更進一步使用ZView 2軟體模擬出感測器之等效電路。未來可配合讀出電路使其最終達到攜帶方便、快速及時得知量測分析結果、直觀、可居家照顧、預防與監測大腸直腸癌之目的。

Since 1981 to 2010, colorectal cancer has become the third Taiwan leading cause of death which rose from the fifth place. During 2010, it was the third major cause of cancer death. Moreover, it placed the fourth cancer death in the world. An advanced biological sensor is proposed in this thesis. This sensor can immediately detect colorectal subject matter (CEACAM6) by its impedance changes. It is more simple, lower cost and time-saving in comparison to other traditional detection methods. Those advantages achieve the goal of early disease discover and early treatment. With screen printing technology, the silver plastic lines and carbon plastic protective layer are printed on the sensor substrate together. Next, CEACAM6 antibody fixed on the biosensor can be fabricated. Measurement results show that the biosensor performs good linearity of R^2 > 0.9 at 610.4Hz ~ 51.76kHz. In this frequency range, it presents the best value of R^2= 0.9978 at 37.35kHz. Furthermore, program ZView 2 is used to simulate the equivalent circuit of this sensor. Eventually, the proposed sensor will be capable of connecting readout circuit to realize ultimately portable, fast and timely detection device. And it can apply to monitoring colorectal cancer for home care and prevention in the future.
