射干化學成分之研究

摘要


射干( Belamcanda chinensis )為鳶尾科( Iridaceae )植物之乾燥根莖,具有清熱解毒、祛痰止咳、活血化瘀的功能。目前射干已被證實具有抗發炎、抑制皮膚真菌、抗病毒、類雌激素(estrogen-like)作用、可誘導人類血癌細胞株(HL-60)細胞分化、清除自由基、抗過敏等生物活性。因雌激素分泌減少,使停經期婦女容易發生更年期症候群,包括代謝性症候群、心血管疾病、骨質疏鬆之機率增加。本研究的目的在於探討射干萃取物及其純化物是否具類雌激素的作用,用以評估射干在骨質疏鬆症藥物開發上的可行性。 以MCF-7細胞株為體外試驗模式,發現甲醇抽取物在10~ 100 μg/ml有促進MCF-7細胞增生作用,顯示其成分可能含有類雌激素作用。從射干粗萃物中分離得到16個化合物,經由光譜分析確認其結構分別為belamoridin (1)、tectoridin (2)、tectorigenin (3)、irisolidone (4)、irilon (5)、irisflorentin (6)、Irilin D (7)、iristectorigenin A (8)、iristectorin A (9)、5,6,7,3'-tetrahydroxy-4'-methoxy-isoflavone (10)、iridin (11) 、p-hydroxybenzaldehyde (12)、kaempferol-3-methyl ether (13)、4-hydroxy-3-methoxyacetophenone (14)、androsin (15)及hydroquinone (16),其中化合物 (1) ~ (12) 為isoflavonoid類化合物,belamoridin (1)為新化合物。將上述isoflavonoid類化合物進行MCF-7之增生試驗,結果發現化合物 (4)、(5)、(6)、(10)在0.001 ~ 0.1 μM濃度下均具有促進MCF-7細胞增生之能力,推測可能具有類雌激素作用。

Belamcanda chinensis (Iridaceae family) is a perennial herb. The rhizome of B. chinensis was used to clear heat-toxin, relieve cough, expelle phlegm, remove blood stasis and resolve static blood in Chinese medicines. B. chinensis have been reported that showed anti-inflammatory, anti-fungal, anti-virus, estrogen-like, scavenging free radical, anti-allergic activities and induced differentiation of human promyelocytic leukemia (HL-60) cell. Due to a decrease in estrogen production, women frequently suffer from menopausal syndromes, including metabolic syndrome, cardiovascular disease and osteoporosis at high risk. The aims of this study were to explore the potential estrogen-like effect of B. chinensis extract and its active ingredients. The methanol extract of B. chinensis showed the proliferation effect on human breast cancer cell (MCF-7 cell). Investigation on the methanol extract of B. chinensis has led to the isolation of a new compound, named as belamoridin (1), along with fifteen known compounds, tectoridin (2)、tectorigenin (3)、irisolidone (4)、irilon (5)、irisflorentin (6)、Irilin D (7)、iristectorigenin (8)、iristectorin A (9)、5,6,7,3'-tetrahydroxy-4'-methoxy-isoflavone (10)、iridin (11) 、p-hydroxybenzaldehyde (12)、kaempferol-3-methyl ether (13)、4-hydroxy-3-methoxyacetophenone (14)、androsin (15) and hydroquinone (16). Their structures were elucidated by spectroscopic evidences. The isoflavonoids, isolated from B. Chinensis, were evaluated for their cell proliferation effect on MCF-7 cells. Compounds 4, 5, 6, and 10 can exhibited the proliferation effect on MCF-7 cells.
