Phostesin是從我們研究中所找出的一個全新PP1結合蛋白,此蛋白質是由KIAA1949所轉譯出來。Phostensin含有一個PP1的結合序列和一個肌動蛋白結合序列分別在胺基酸91-94以及125-155的位置上。然而從本實驗室研究中發現phostensin可以結合在肌動蛋白絲的pointed end上並影響肌動蛋白的動態平衡。雖然我們已經鑑定了phostesin其生化功能,但在生物上的功能卻仍然不清楚 , 因此需要準備phostensin缺失老鼠並分析phostesin在生物功能上的意義。 同時,老鼠phostesin的抗體對於我們研究上是必須的。此外我們也想知道phostensin在其他物種中的生化功能是否也與人類相似。所以,我的實驗是建構並純化老鼠phostensin蛋白質,而其將會被用於準備老鼠phostensin抗體和分析其生化功能。
Phostensin is a protein phosphatase 1 (PP1)-binding protein. This protein is encoded by KIAA1949. Phostensin consists of 165 amino acids and contains one consensus PP1-binding motif at residues 91-94 and one actin-binding motif at residues 125-155. Phostensin can cap the pointed end of actin filament and modulate the actin dynamic. Although we have identified the biochemical function of phostensin, its biological functions remained unknown. Therefore, we have carried out to prepare phostensin knockout mice and analyze the biological functions of phostensin. In the meantime, the anti-mouse phostensin antibody was required for our studies. In addition, we also want to know the biochemical characteristics of phostensin in other species. Thus, my experiment is to clone and prepare the recombinant mouse phostensin. The protein will be used for preparation of mouse phostensin antibody, and analyze it biochemical function.
