三裂葉蟛蜞菊花萃取物於減緩肝癌形成及其安全之評估

摘要


三裂葉蟛蜞菊(Wedelia trilobata (L.) Hitchc),又稱南美蟛蜞菊。民間記載具有清熱、解毒之功能,亦常用於植披及美化環境。本研究以三裂葉蟛蜞菊之花為原料,經均質、酒精萃取及濃縮後以安氏實驗(Ames test)評估其安全性,判定對於各測試菌株並無毒性及致突變影響。且在抗致突變試驗中,以最高濃度5 mg/plate測試時,對於市售致突變藥物4-NQNO所造成之致突變現象具27.9% ~ 68.1%之抑制能力。另以動物試驗探討萃取物對於大鼠體內肝癌結節產生之延緩效果。以添加0.01% Diethynitrosamine (DEN)於動物飲水中進行肝癌誘導,每日以管餵方式給予萃取物,並與市售藥物Silymarin作為對照組。結果顯示給予最高劑量(100 mg/Kg B.W.)之安全組體重和控制組相比並無顯著差異,分別為447.9±9.0 g及487±21.0 g。且GOP及GPT指數為47.8±10.8 IU/L、47.6±9.3 IU/L及30.4±9.2 IU/L、28.4±6.5 IU/L,判斷在100 mg/Kg B.W.之萃取物對大鼠應無不良影響。在實驗組別則以中劑量組別(50 mg/Kg B.W.)最具有顯著性效果,GOT及GPT相較於負控制組分別下降了21.6%及20.3%,且GSH及CAT之活性之降低幅度亦為組別間最低。然給予10 mg/Kg B.W. Silymarin並無法明顯減緩GOT及GPT之上升現象,僅在SOD活性有稍微延緩下降之現象。推判原因為劑量濃度不足,無法延緩肝癌形成。

Wedelia trilobata (L.) Hitchc, a kind of herb, it demonstrated a good ability on detoxification. The aim of the present study is to evaluate the safety of ethanol extract from Wedelia trilobata flower by Ames test. Little toxicity and mutagenicity were found, the antimutagenicity ability was 27.9% ~ 68.1% toward S. typhimurium TA 98, TA 100, TA 1537, TA 1535 and E. coli WP-2. In animal study, 0.01% diethylnitrosamine mixed with drinking water was employed for inducing liver tumor. The results suggested that the extract of Wedelia trilobata didn’t show significant effect on experimental subjects, Sprague-Dawley rats, according to the body weight data and acute toxicity animal results. The body weight for control and sample group is 447±9.0 g and 487±21.0 g, respectively. Moreover, GOT and GPT for control and sample group is 47.8±10.8 IU/L、47.6±9.3 IU/L and 30.4±9.2 IU/L、28.4±6.5 IU/L, respectively. Conclusively, the results suggested that the middle dosage (50 mg/Kg B.W.) indicated the best effect, whereas the GOT and GPT value decreased 21.6% and 20.3% compared to negative group, and inhibit the decrease of GSH and CAT. At silymarin group, which the dosage is 10 mg/Kg B.W, could not exhibit good ability on decrease of GOT and GPT due to the low dosage.
