臺灣地區第三型豬環狀病毒之調查及序列分析

摘要


豬環狀病毒 (Porcine circovirus, PCV)屬於環狀病毒科,環狀病毒屬,為具有環形單股DNA病毒。豬環狀病毒現在有第一型豬環狀病毒 (Porcine circovirus type 1, PCV1)、第二型豬環狀病毒 (Porcine circovirus type 2, PCV2)及第三型豬環狀病毒 (Porcine circovirus type 3, PCV3)。其中第二型豬環狀病毒,造成養豬界嚴重的經濟損失。近年來有研究,在有些豬皮膚炎與腎病症候群 (Porcine dermatitis and nephropathy syndrome, PDNS)及繁殖障礙 (Reproductive failure, RF)病例中,利用即時聚合酶鏈鎖反應方法並未檢測到PCV2。但藉由組織病理學配合分子生物診斷技術,則檢測到一個新型的豬環狀病毒,被命名第三型豬環狀病毒。目前在臺灣並無第三型豬環狀病毒的研究,因此本研究之目的在於檢測臺灣是否有PCV3的存在。本研究收集 2016 年 1 月至 2018 年 3 月的保育豬、母豬和流產胎之共1712個樣本。其第三型豬環狀病毒的樣本陽性率分別為保育豬7.4% (85/1195)、母豬3.1% (3/94)和流產胎4.0% (17/423)。此結果證實第三型豬環狀病毒存在於臺灣。為了更進一步探討臺灣第三型豬環狀病毒與其他國家序列的關係,我們進行全長基因、ORF1 (Open reading frames1, ORF1)及ORF2 (Capsid protein gene, cap gene)的親緣性之比對,顯示臺灣的PCV3病毒株與各國的PCV3病毒株極為相似。第三型豬環狀病毒在保育豬當中檢出率最高,而PCV3也與其他病原共同感染,因PCV3的致病機轉尚未明白,未來仍須透過實驗研究來確定PCV3與臨床之關聯。

Porcine circovirus (PCV) belongs to the Circoviridae family, and Circovirus genus. It is a single strand circular DNA virus. Now porcine circovirus have Porcine circovirus type 1 (PCV1), Porcine circovirus type 2 (PCV2) and Porcine circovirus type 3 (PCV3). So far porcine circovirus type 2 has caused great economic losses in the swine pig industry worldwide. Recently, there have been some cases of porcine dermatitis and nephropathy syndrome and reproductive failure and PCV2 detection was negative by the Real-time polymerase chain reaction (RT-PCR). However, PCV3 was detected by histopathological and molecular biological tests. There is limited information regarding PCV3 infection in Taiwan. Thus, the aim of this study was to analyze PCV3 in Taiwan. A total of 1712 specimens from nursery pigs, sows, and were collected from January 2016 to March 2018. PCV3 positive rates were 7.4% (85/1195) for nursery pigs, 3.1% (3/94) for sows and 4.0% (17/423) for abortion fetus. These results confirmed the presence of PCV3 in Taiwan. Investigate relations by further Taiwanese PCV3 and other countries, sequence and phylogenetic analyses suggest that the sequence of PCV3 the complete genome, ORF1 and ORF2 are similar to those from other countries. The highest detection rate of PCV3 samples were nursery pigs. PCV3 may co-infection with other pathogens. The pathogenesis of PCV3 is still unclear, clinical observation in the field is needed by using molecular biology detection and experimental study to determine the association between PCV3 and clinical manifestations.
