膽固醇是對人類生活是至關重要的親脂性分子,它具有許多有助於細胞正常運作的作用,然而過多的膽固醇則會造成高膽固醇血症,進而增加許多疾病的罹患風險,包含腦血管疾病、心臟病、糖尿病、高血壓等,因此膽固醇的測定及研究顯得格外重要。 常見的相關研究大多選擇簡單易操作且快速的市售免疫分析套組(Enzyme-linked immunosorbent assay, ELISA),本研究則是選擇高效液相層析 (High performance liquid chromatography, HPLC) 與其同步分析,比較其兩者差異性,結果為市售分析套組在雄性敘利亞黃金倉鼠之肝臟與糞便檢體皆顯著的高於高效液相層析分法;然而在以紫檀芪 (Pterostilbene, Pt) 及其衍生物3'-hydroxypterostilbene (OHPt) 處理脂質累積的HepG2細胞膽固醇含量測定卻得到相反的結果。 雖然高效液相層析法靈敏度、準確度及特異性皆優於市售分析套組,但由於需要許多複雜的操作參數,若是不合適的處理或條件皆可能影響結果,因此可針對樣品進一步探討並優化條件是必要的。 關鍵字:膽固醇、高效液相層析、市售酵素免疫分析套組
Cholesterol is a lipophilic molecule that is essential for human life. It has many roles that contribute to normally functioning cells. However, too much cholesterol in the body could cause hypercholesterolemia. It increases the risk of many diseases, including cerebrovascular disease, heart disease, diabetes, high blood pressure, etc. Therefore, it is particularly important to choose a suitable cholesterol analysis method. The most of research chose the commercial kit, because it is easy to operate and fast. This study used high-performance liquid chromatographic analysis (HPLC) to compare the difference with commercial kit. The results showed that the cholesterol concentrats of liver and feces of male Syrian hamster measured using commercial kit is higher than those using HPLC. But we got the different results in the samples of pterostilbene (Pt) and its derivative 3'-hydroxypterostilbene (OHPt) treated the lipid accumulation’s HepG2 cells. However, this results can be reliably explained in other studies that use these two analytical methods for comparison. Although the sensitivity, accuracy and variability of high-performance liquid chromatography are better than commercial analysis kits, but due to the need for many complex operating parameters. If use inappropriate processing or conditions may affect the results, so further target the sample to explore and optimize the conditions is necessary.
