五倍子甲醇萃取物溶劑區分之抗氧化性質

摘要


五倍子為漆樹科植物鹽膚木 (Rhus chinensis Mill.)葉上的乾燥蟲 蠳,為民間常用之中藥,具有抗潰瘍、抗凝血、抗菌、前列腺素合成 抑制等藥理作用。本研究探討不同溶劑對其抗氧化能力之影響,並利 用溶劑區分對五倍子具抗氧化活性之成份進行初步分離。結果顯示以 80%乙醇萃取五倍子之萃取率最佳,為87.16%。80%乙醇萃取物之 DPPH自由基清除率較80%甲醇及熱水萃取物者高。還原力及螯合亞 鐵離子能力則以80%甲醇萃取物較佳,其螯合 50 % 亞鐵離子力濃度 (EC50) 為 2.599 mg/ ml。熱水萃取物之總酚含量最高,為797.0 mg gallic acid / g。五倍子甲醇萃取物經過n-hexane 、diethyl ether 、ethyl acetate、1-butanol 及水區分後,進行各區分層之抗氧化活性探討,發 現ethyl acetate區分層之DPPH自由基清除率及還原力最高,螯合亞 鐵離子能力則以水區分層最佳。總酚含量則以ethyl acetate區分層最 高,每克之區分物中含有504.8 gallic acid。

Galla rhois, the dry gall on the leaves of Rhus chinensis (Anacardiaceae), was used in traditional Chinese medicine. It exhibited some pharmacological properties, including anti-ulcer, anti-clotting, antibacterial and repression of prostaglandin synthesis. The effects of solvent varieties on the antioxidant activity of Gall rhois and antioxidant capacity-guided fractionation were conducted in this study. The results showed that the extraction yield, ca. 87.16 %, was the highest when 80% ethanol was used as extraction solvent. DPPH free radical scavenging activity of 80% ethanol extract was higher than those of methanol extract and hot water extract. The methanol extract showed demonstrated the highest reducing power and ferrous ion chelating activity. The efficient concentration of chelating 50% of ferrous ions (EC50) was 2.599 mg/ ml. The highest content of total phenolic, ca. 797.0 mg gallic acid / g, was found in hot water extract. Methanol extract was partitioned sequentially with n-hexane, diethyl ether, ethyl acetate, 1-butanol and water and then the antioxidant activity of each fraction was determined. The DPPH free radical scavenging activity and reducing power were highest in ethyl III acetate fraction. On the other hand, the water fraction exhibited the highest ferrous ion chelating activity. The highest phenolic content, expressed as 504, 8 mg of gallic acid per grem of fraction, was obtained in ethyl acetate fraction.
