探討BK 病毒次要結構蛋白在病毒繁殖過程中扮演的角色

摘要

BKV 和 JCV 屬於人類多瘤性病毒都屬於多瘤性病毒家族的成員， BK 病毒是造成腎臟功能損壞的重要原因，常見於愛滋病患者或是免疫力低下的病人。在人類多瘤性病毒 BKV 的病毒殼體內，包含有結構蛋白 VP1、VP2、VP3 和病毒的 DNA，在病毒的生活週期中扮演很重要的角色。同家族中的 JCV 其立體結構目前已被解析出來。JCV 是由 72 個 VP1 的pentamers 所組裝而成，此 pentamers 稱之為 capsomere。由 X-ray 繞射所得到結果觀察得知每個 pentamer 中心凹洞有一個次要結構蛋白 VP2 或VP3 的存在。
JCV 和 BKV 的 VP2 和 VP3 都有 DNA binding domain、nuclear localization signal 和 VP1-interaction domain 。但到目前仍不清楚次要結構蛋白 VP2 和 VP3 在病毒殼體的組裝及生理上，各扮演什麼角色。在我的研究中，我們發現 VP2 和 VP3 在病毒複製週期是必需的。本研究中將病毒基因 VP2 和 VP3 knockout，送到 vero cell (猴腎細胞) 大量表達，並利用 Western blot 及 IFA 技術觀察，發現突變的 BK 病毒無法進行早期轉錄的功能進而也影響到病毒無法產生病毒子代。

The human polyomavirus, BK virus (BKV) and JC virus (JCV), were the members of family polyomaviridae. BK virus produces a mild respiratory infection and affect the kidneys of immunosuppressed transplant patients. The human polyomavirus, BK virus, contains three capsid protein, VP1, VP2, VP3 and a viral minichromosome. Interactions of these three capsid proteins for virus assembly were not well understood.
In this studies, Collect lysates from vero cell (3x105) that had been transfected with BK mutant VP2 or VP3, detected by Western blotting and Immunoflourescence Assay. To find out the effect of proteins expression of BK VP2 and VP3 mutants.
All these results suggest that BK VP2 and VP3 mutants can effect the expression of virus early genes and late genes, lead to the decreasing of propagation ability. In this study, we found VP2 and VP3 are both required for virus propagation.
