利用液相層析圖譜相似性分析方法探討市售芍藥藥材

中文摘要 :在現代人追求健康的風潮下,中藥雖然被重新發現其價值,但藥材品質的優劣,才是真正影響醫療成果之重要關鍵,所以如何穩定藥材的品質,建立品質評價資料等工作,誠屬重要之課題。中藥指紋圖譜已成為研究中藥製劑乃至世界各國植物藥製劑的物質基礎和作用機制的一種新模式研究體系。芍藥(Paeoniae Radix)為中醫臨床上常用之清熱解毒、養血調經之中藥,現代藥理實驗更證明芍藥的主要成分芍藥苷(Paeoniflorin) 具有鎮痛、抗發炎及降低血栓素的合成,改善血液循環等作用。故本研究建立市售的芍藥藥材層析圖譜,運用多變量統計分析,探討芍藥品質的穩定性,找出一個相似性分析客觀的評估方法,使中藥能夠標準化,進而達到中藥鑑定的工作。 研究結果顯示台灣北、中、南、東等地市售芍藥藥材所獲得層析圖譜的重疊率與共有峰率值互有高低,此乃藥材複雜性較高所致。不同的萃取方法利用群聚分析、主成分分析與多樣性指數所計算出的結果也不同,而南部地區的芍藥藥材相似度相對於其他區域為低,此表示芍藥藥材複雜性較其他區域高。

Abstract :In the wave of pursuing healthy life for the modern society, Chinese medicines have been rediscovered its value. It is believed that the quality of the crude drug material of these medicines is actually the key for a successful medical treatment. Therefore to establish a system for evaluating the quality of those herb medicines becomes a very importance work. The Chinese herbal medicine fingerprinting map has already become a new model to study the products of Chinese herbal medicine for understanding the material foundation as well as the function mechanism. Meanwhile using this technique can also apply to the international community for all kinds plant medicine products. Paeonia Radix is one of the most important Chinese medicine herb used mainly in treating diseases such as inflammation, allergy and some women’s diseases. The recent pharmacology studies proved that the Paeoniflorin, the main composition of the Paeonia Radix, have the functions of ant-inflammation, antiallergic and promoting blood circulation to remove blood stasis. In this study, a chromatogram recognition of main chemical constituents of Paeoniae Radix collected from different cities was used in the multivariate analysis to evaluate the quality stability of Paeoniae Radix. It is thought that through this similarity analysis of Paeonia Radix crude drugs could offer the basis for quality control in a standard way. Our result shows that the Paeoniae Radix sold in different cities in Taiwan have different characteristics with overlapping and common peak ratio in their chemical chromatogram maps and this is assumed because of the high complexity of Paeoniae Radix itself. Using cluster analysis, principal component analysis and the multiple index for the different extract methods indicated a diverse results. A lower similarity is found for the materials purchased from the southern areas of Taiwan than that of taken from other areas in Taiwan. Again, this shows a higher complexity of crude drugs taken from this areas.
