免疫組織化學染色(immunohistochemistry, IHC)是利用抗原和抗體間特異性結合反應,並透過螢光或可呈色化學顯色劑檢測細胞或組織中是否有目標抗原的存在。經由IHC可以在生物實驗上驗證生物晶片(Microarray)所提供的致癌基因或抑癌基因在生物樣本表現的反應是否與其結果一致。生物晶片通常用來檢測或分析生物性分子,由於反應快速且能夠平行分析大量生物資訊,因此適用於疾病檢驗、基因表達和新藥開發等...等。雖然透過生物晶片這個工具能夠快速的得到大量致癌基因與抑癌基因的資料。由於IHC影像在傳統上都是利用人力去一張一張作判讀,速度不僅過慢且由於在部份IHC影像上,正常組織與癌化組織並沒有明顯的分隔線,人力上很難去作判讀。 因此我們開發出,一個自動化分析IHC影像的系統工具,這個工具最主要的目的有二點:1. 利用電腦協助判斷影像的正常組織與癌化組織的分佈;2. 利用自動化判定標記染色蛋白在正常組織與癌化組織的表現量差異。 而透過本系統,除了能夠快速的得到IHC影像上正常組織與癌化組織區域判讀外,能藉由瞭解特定蛋白分佈在此兩區域的表現量,得知基因與癌症上的關係。藉此輔助生物晶片上所得到的結果作證明。
