地耳草大量繁殖之研究

中文摘要 地耳草(Hypericum japonicum Thunb.ex Murray)為金絲桃科(Guttiferae)、金絲桃屬(Hypericum)一年生草本植物,本品始載於植物名實圖考。地耳草具有抗菌消炎、抗瘧、預防傳染性肝炎、胃腸疾病和抑制腫瘤等作用。現代藥理研究具有治療急、慢性肝炎。 本省因土地開發及利用而破壞其生育地,為開發省產地耳草之生藥資源並解決地耳草生態保育問題及產業發展,本研究將野生地耳草莖段培養於含有 2 mg/l BA、0.2 mg/l NAA之 MS基本鹽類培養基中,初誘導出大量不定芽,供本研究探討各種不同因素影響之用。實驗結果顯示地耳草莖段誘導芽體增殖以含有2 mg/l Kinetin之1/2MS培養基可獲得10.2個芽體。若改將芽體培養於含有2 mg/l Kinetin及 5% 蔗糖之1/4MS培養基中,可得26.1條不定根。 利用組織培養方法培養地耳草,不僅可克服土地、氣候及環境因素變化的限制,並可藉由建立地耳草組織培養以達到快速繁殖技術之目的。

Summary Hypericum japonicum Thunb. ex Murray, commonly called as “Di-Er- Cao”in Taiwan , is a species of Hypericum , Guttiferae genera , it was first recorded in Chin- Wu-Ming-Shih-Tu-Kao, has been as a Chinese herbal medicine for the treatment of several bacterial diseases, infectious hepatitis, gastrointestinal disorder, internal hemorrhage and tumors. The pharmaceutical industry is mainly dependent upon wild populations for supplies of the plant material. Due to large-scale and over-harvesting of wild material from forests, H. japonicum is rapidly disappearing. The purpose of this study is to establish a tissue culture technique for the mass propagation of H. japonicum. In the study we found that the most efficient shoot formation was the nodal stem explants, cultured on MS medium supplemented with 2 mg/l BA, 0.2 mg/l NAA, then transferred onto solid medium containing 1/2 MS basic salts with 2 mg/l Kinetin, 1.0% agar and 3% sucrose, forming 10.2 shoot per explants the bast medium for rooting was 1/4 MS basic salts with 2 mg/l Kinetin and 5% sucrose, gave the highest root number of rooting. forming 26.1 root per explants. Using tissue culture method of raise H. japonicum, not only to overcome the land, climate and environmental factor change limit, and by the establishment of tissue culture to H. japonicum rapid propagation techniques in order to achieve the purpose.
