光化學藥物Hematoporphyrin加光照射對口腔癌細胞株之毒殺力之研究

摘要


Hematoporphyrin derivative (HPD)是一種光化學藥物,此藥物在螢光下會發出紅光,若經可見光照射即可變爲活性,放出singlet O2殺死癌細胞;目前已做爲檢查和追蹤癌細胞以及治療癌症之用。 本篇報告是測試HPD在體外對口腔上皮樣癌細胞株之殺傷力以及比較對其它癌細胞株之敏感度。約5×10^4 OC1及OC2口腔上皮樣癌細胞,分別加入10μg,5μg,2.5μg,1.25μg,0.63μg/ml之HPD,一小時後沖洗乾淨,再用630nm的可見光照射,然後用AO/EB螢光染色法來測量其毒殺力。結果發現5μg/ml的HPD就幾乎可以完全殺死兩株口腔上皮樣癌細胞株;如果只加HPD而沒有加光線照射,細胞不會死亡;若不加HPD而只以630nm光線照射一小時,癌細胞也不會被毒殺。兩種口腔上皮樣癌細胞株OC1,CO2對HPD的敏感性略比肝,肺癌細胞株差,比口腔正常黏膜上皮細胞敏感度稍高。HPD對於口腔癌之治療是深具潛力的一種藥物。

Hematoporphyrin derivative (HPD) is a photochemical drug. It has a selective affinity for malignant cells and it can also be activated by visible light to kill the tumor cells. Its cytotoxic effect is mediated through the production of singlet oxygen. In this study the cytotoxicity of the HPD to human epidermoid carcinoma cell lines, human hepatoma cell line, human lung cancer cell line was observed. HPD at concentration of 10, 5, 2.5, 1.25, 0.63 and 0μg/ml was added in cultured cells and irradiated with 630nm red light. The cytotoxicity was measured by Acridine Orange Ethidine Bromide. The results showed that HPD in such low doses has no killing effect by itself, but 5μg/ml HPD irradiated with 630nm red light can kill almost all human epidermoid carcinoma cell lines. A higher dose of HPD was required to kill the OC1 and OC2 cell lines than it was required to kill the hepatoma cell line and lung cancer cell line. A slight differentiation in susceptibility was seen between carcinoma and normal gingiva epidermoid cells.
