天麻的抗癲癇作用與Interleukin-1β、Tumor Necrosis Factor-α及一氧化氮關係

摘要


目的 我們先前的研究已知天麻能減少kainic acid(KA)誘發大鼠的癲癇發作和抑制microglia的活化,因此本研究目的是探討天麻的抗癲癇作用與interleukin-1β(IL-1β)、tumor necrosis factor-α(TEN-α),及一氧化氮(nitric oxide:NO)的關係。 方法 使用KA 12mg於Sprague-Dawley(SD)大鼠腹腔注射誘發癲癇發作,觀察天麻1.0g/kg、0.5g/kg,以及Valproic acid(VA)250mg/kg前治療四天對濕狗甩頭(wer dogshakes),血液中IL-1β,血液中的IL-1β、TNF-α和NO的濃度,以及前額葉與海馬區IL-1、α、TNF-α濃度的影響。 結果 天麻1.0 g/kg和VA 250mg/kg能減少KA誘發濕名狗甩頭次數和周邊血液IL-1β和一氧化氮的濃度,以及鼠腦前額葉IL-1β、TNF-α和海馬區TNF-α的濃度。 結論 天麻的抗癲癇作用抑制細胞因子IL-1β、TNF-α的濃度。

Purpose Our previous studies have shown that Gastrodia elata (GE) reduces kainic acid (KA)-induced epileptic seizures and inhibits activation of microglia in rates. The present study investigated the relationship between the anticonvulsion effects of GE and interleukin-1β (IL-1β), tumor necrosis factor-α (TNE-α), as well as nitric oxide (NO). Methods Rats were administered KA (12mg/kg) to induce epileptic seizure. After oral administration of GE 1.0g/kg, 0.5g/kg and valproic acid (VA) 250mg/kg for 4 days prior to K injection of the number of KA-induced we dog shakes was calculated, and the changes in IL-1β, TNF-α and NO levels in the peripheral blood, and IL-1β and TNF-α levels in the frontal cortex and hippocampus regions of the rat brain were measured. Results Both pretreatment of GE 1.0g/kg and VA 250mg/kg reduced the number of wet dog shakes. GE also reduced the levels of IL-1β and NO in the peripheral blood, and the levels of IL-1β and TNF-α in the frontal cortex, and TNF-α in the hippocampus. Conclusions GE inhibits the generation of cytokine IL-1β, TNF-α, and NO in rats with KA-induced seizures.
