蘿蔔嫩莢(Silique Raphani)之急性毒性試驗

摘要


本論文探討蘿蔔嫩莢對小白鼠之急性毒性評估,採用Litchfield and Wilcoxon方法,分正常組及實驗組(蘿蔔嫩莢水抽物劑量:0.0mg/kg、1,000mg/kg、3,000mg/kg、10,000mg/kg),總共四組。進行口服單劑量之短期試驗,觀察小鼠死亡數目及中毒情形以計算急性毒性之半數致死劑量(LD50)。此外,觀察期結束後採取小鼠的血液及臟器,分別檢測血清生化指標及組織病理切片。結果:急性毒性試驗中,四組皆無死亡,其半數致死量為每公斤大於10.0公克,顯示蘿蔔嫩莢之水抽物毒性甚小。生化指標探討在肝功能和腎功能方面:與對照組並無明顯差異;在代謝方面:BUN和膽固醇有減降的趨勢且在高劑量達統計差異。在組織切片上,實驗組與正常組一樣,並沒有特殊的病變表現,各臟器正常。

The evaluation of acute toxicity of “Si1ique Raphani” on mice was based on the Litchfield and Wilcoxon method. All mice were divided into four groups, one control group and three experimental groups (dose: 0.0mg/kg, 1, 000mg/kg, 3, 000mg/kg, 10, 000mg/kg). Each dose of the crude extract of “Si1ique Raphani” and normal saline was administered oral 1 y to those mice. All the animals were observed 72 hours successively to evaluate their toxicological responses and mortality and thus calculate the semi-lethal dose (LD50). After 72 hours, all animals were sacrificed and their blood and target organs were collected. The collected sera were examined to determine the biochemical function, including liver functions (GOT, CPT), nephritic function (BUN, Cretonne) and metabolic function (cholesterol, glucose). Besides, histological slides were performed to evaluate their pathological change. The experiment results indicated the crude extract of “Silique Raphani” is extremely slightly toxic (LD50> 10. 0g/kg). It didn’t cause any death in mice. In fact, the experimental groups did not show any significant difference in serum biochemical data and pathological change in slides if compared to the control group.
