針刺足三里對先天性高血壓大白鼠心肌細胞的影響及其基因表現探討

The Effects and the Gene Expression of Acupuncture on Cardiac Muscle Cells in Spontaneous Hypertensive Rat

摘要


本篇旨在探討針刺先天性高血壓大白鼠是否可減緩心肌肥大,進一步分析心肌細胞之基因表現,進而抑制細胞凋亡。發現針刺治療後,心臟重量變化及其與體重的比值變化雖不明顯。但針刺六天後在足三里(ST.36)穴位組發現,大白鼠心肌細胞大小顯著較小,改善了先天性高血壓大白鼠心肌細胞肥大的現象。針刺三天穴位組其訊息蛋白表現中,大部分訊息蛋白的表現變化不明顯但針刺六天穴位組,除與心肌remodeling及抑制纖維化之matrix-metallo-protease-2((MMP-2)比先天性高血壓大白鼠顯著的恢復上升之外,與心肌細胞存活(Survival)有關之 IGF-I、Bcl-2、phosphatidylinostol-3 ( PI3-kinase[(PI3K)]及磷酸化Bad ( Bad-P)等抗細胞凋亡蛋白含量明顯增加。而caspases3、caspasesg、Caspasess等促凋亡酵素則明顯下降,惟Fas、FADD等Death Receptor無明顯改變。研究顯示針刺三天穴位對心肌細胞存活調控之訊息蛋白表現影響較不明顯,但是針刺六天穴位組,除心肌細胞肥大現象改善外,與心肌細胞存活有關的訊息蛋白量則明顯改變。實驗數據指出針刺抗心肌細胞凋亡乃是透過Survival Factors ( IGF-I)及其下游相關途徑所促進。相信採用適當且足期的針刺可減緩基因缺陷引起的心肌肥大現象及抑制心肌細胞凋亡。

In this research, the aim is to discover whether the acupuncture stimulation can reduce the hypertrophy of the heart and stop apoptosis of the cells. In this experiment, the heart weight and size of the heart muscles have no significant changes. After 6 days of needling on the Zusanli (ST.36) points, the size of the myocardial cells is significantly reduced and the hypertrophy of the heart in the spontaneous hypertensiue rats is also improved. In the treatment groups, after 3 days of needling, there are no any significant changes in most of the message transmission proteins. However, after six days of needling, the cardiac muscle remodeling and the fibrosis suppressor, matrix-metallo-protease-2 (MMP-2), have significant recovered and rose when comparing to the spontaneous hypertension rats. The proteins that related to the myocardial cells survival rate, the anti-apoptosis proteins, such as IGF-I, Bcl-2, phosphatidylinostol-3-kinases (P13K) and phosphate Bad (Bad-P) are all increased. On the other hand, the apoptosis inducing enzymes, Caspases3, Caspases9 and Caspases8, are all reduced. In addition, the death receptors, Fas and FADD, have no any significant changes. Research studies show that the Zusanli (ST. 36) points, after needling for 3 days, the transmission proteins, have no any significant changes. However, after 6 days of treatment, the condition of the hypertrophy of the heart had improved, and the myocardial cells survival controller, the transmitter proteins, also had significant changes. From the laboratory data indicates that acupuncture needling can defend the apoptosis of the inyocardial cells under survival factors (IGF-I) and its related pathways. Therefore, in the clinical treatment, I believed when acupuncture needling is applied with sufficient period of time, it can reduce the hypertrophy of the heart that caused by defect genes and suppress apoptosis of the myocardial cells.
