薑黃素結合遠紅外線可降低在THP-1細胞經PMA誘導的MMP-2和MMP-9活性

Curcumin and Far Infrared Radiation Can Reduce PMA-induced MMP-2 and MMP-9 Activities in THP-1 Cell

摘要


薑黃素(curcumin)擁有調節細胞訊息傳遞的途徑(NFκB、Bax)進而抑制癌細胞的增長,薑黃素也能調控MMP-2和MMP-9發炎反應的活性表現量,文獻記載指出遠紅外線可有效抑制腫瘤細胞的增長,遠紅外線也能治療發炎相關疾病。本研究探討薑黃素與遠紅外線結合對人類單核球細胞的生物效應之研究。使用薑黃素試劑在不同薑黃素濃度分別爲1μM、5μM、10μM,再經由遠紅外線(far infrared)儀器距離培養血30公分,分別照射不同時間5分鐘、10分鐘、15分鐘,經過24小時作用時間,收集細胞外的培養液,利用Zymogrphy assay來測試MMP-2和MMP-9活性表現量。研究結果顯示出薑黃素對MMP-2活性的表現量並不顯著,分別爲98%(1μM)、101%(5μM)、99%(10μM),因此我們添加100μM/ml PMA(phorbol-12-myristate-13-acetate,可消化protein kinase C誘導細胞大量表現MMP-2和MMP-9)誘導MMP-2和MMP-9活性大量表現作用時。再加上述不同濃度的薑黃素結果對MMP-2活性表現量也隨著薑黃素濃度增加而受到明顯的抑制,其抑制效果分別爲90%(1μM)、80%(5μM)、79%(10μM)。另一方面,遠紅外線研究結果所顯示出在單獨使用不同時間照射時,對MMP-2活性表現量並不顯著分別爲99%(5分鍾)、102%(10分鐘)、98%(15分鐘),因此我們添加100nM/m PMA誘導MMP-2和MMP-9活性大量表現作用時,再加上述不同遠紅外線照射時間結果對MMP-2活性表現量也隨著遠紅外線照射增加而受到明顯的抑制。其抑制效果分別爲89%(5分鐘)、86%(10分鐘)、78%(15分鐘)的百分此。其結果顯示在單獨添加薑黃素和遠紅外線照射時,對MMP-2和MMP-9活性並不顯著,另外,當添加100nM/ml PMA誘導MMP-2和MMP-9活性時,薑黃素和遠紅外線照射時,具有顯著抑制MMP-2和MMP-9活性。

The curcumin possess the ability to regulate cell signal transcription pathways (nuclear factor κB; NFκB, Bax) inhibit cancer cells proliferation. It also involves the down-regulation of the activities of matrix metalloprotenases (MMPs) and inflammatory mediators. The FIR has inhibition effects on tumor cell growth and therapeutic functions in autoimmunity and inflammation diseases. This study evaluated the biological effects of both curcumin and far infrared radiation in K562 and THP-1 cells.Curcumin in anti-inflammatory effect, low concentration of curcumin (1 μM, 5 μM and 10 μM) in the activity of MMP-2 was no significant effect. When THP-1 cells stimulated with 100 nM/ml PMA (phorbol-12-myristate-13-acetate, PMA bind to protein kinase pathways induces cells expression MMP-2 and MMP-9). Add on different concentration of curcumin treatment. The activitys of MMP-2 and MMP-9 in the THP-1 cells stimulated with PMA by 90% (1 μM). 83% (5 μM) and 68% (10 μM) curcumin treatment caused significant inhibition and dose dependent effect. In addition, the study found in independent used different exposure time of far infrared (5 minute, 10 minute and 15 minute). Far infrared in the activity of MMP-2 was no significant effect. When THP-1 cells stimulated with 100 nM/ml PMA induced the activity of MMP-2 and MMP-9. Add on different exposure time of far infrared treatment cells. We found the activitys of MMP-2 and MMP-9 by far infrared treatment caused significant inhibition and time-dependent effect.
