白茅根萃取物之生物活性分析

The Study of Anti-cancer and Antioxidant Effects in Rhizoma Imperatae Extracts

摘要


白茅根具有利尿解熱、調解免疫及抗癌的功效。本次研究乃利用白茅根萃取物進行生物活性分析,採取方式爲傳統的熱水萃取法,再利用真空乾燥,取其根部的粉末加入PBS回溶,取實驗細胞株血癌細胞(K562)及肝癌細胞(HUH-7),經不同濃度白茅根萃取液處理後,觀察其細胞存活率及型態。首先做白茅根的抗癌分析,以50μg/ml、100μg/ml、300μg/ml、500μg/ml,加入肝癌細胞血癌細胞中,24小時後觀察其細胞。結果發現,細胞的存活率與控制組相當,白茅根對此兩種細胞沒有抗癌效果。在白茅根的抗氧化分析中,加入HuH-7(細胞株)24小時後,再加入H2O2(Hydrogen peroxide solution 10mM)並在37℃培養24小時後觀察細胞型態存活,與具有抗氧化效果的維生素C(100μM)作爲對照組,觀察HuH-7在氧化壓力下的存活率。實驗發現,在抗氧化實驗中,白茅根在濃度較高(500μg/ml)情況下,細胞比控制組(添加維他命C)和對照組(只添加PBS)細胞存活高。據文獻資料指出,白茅根具有清熱的效果,所以利用西方點墨法來確認白茅根是能誘導細胞內Hsp70蛋白質(Heat Shock protein)的表現,結果顯示,經白茅根處理的的肝細胞Hsp70蛋白質和控制組並無顯著差異。

Rhizoma Imperatae has dieresis, solve heat, immunity regulation and anticancer activities. This research was to utilize Rhizoma Imperatae extracts and carry on the biological analysis. We extracted Rhizoma Imperatae with traditional hot water method, utilized vacuum to be dry and then used PBS buffer to resolve. The extract was then added to a blood cancer cells (K562) and the liver cancer cell (HUH-7) to observe its survival rate and cell morphology. At first, we treated the two cell lines with 50 g/ml, 100 g/ml, 300 g/ml, 500 g/ml, and observed their survival rate and cell morphology 24 hours later. The result showed us that Rhizoma Imperatae extracts did not have anticancer activity to these two kinds of cancer cells.In the antioxidant analysis of Rhizoma lmperatae extracts, HuH-7 cells were pretreated with extracts for 24 hours, and then added H2O2 (Hydrogen peroxide solution 10 mM), in the same time the vitamin C as the control group. Data from this experiment demonstrate Rhizoma Imperatae extracts in 500 g/ml concentration has more antioxidant activity than vitamin C. Finally, we also to analyze whether if Rhizoma Imperatae extracts could regulate the heat shock protein 70 (Hsp 70) expression. However, experimental data showed us that Rhizoma Imperatae had no effect in regulate the Hsp 70 expression.
