香蘭葉水萃取物對肝癌細胞HepG2之存活與凋亡的影響

Effects of water extract of Pandanus amaryllifolius on survival and apoptosis of hepatocellular carcinoma HepG2 cells

摘要


香蘭(Pandanus amaryllifolius)的葉片萃取物已知具有抗氧化、降血糖、抗病毒、抗癌細胞生長與促進細胞凋亡等作用。為探討香蘭葉內的化學物質是否具有肝癌療效,本研究利用超音波萃取法取得香蘭葉水萃取物,分析香蘭葉水萃取物對於人類肝細胞癌之細胞株HepG2 之存活與凋亡的影響,以及其作用的可能分子機制。結果顯示,經香蘭葉水萃取物處理72 小時後HepG2 細胞存活率會被明顯抑制,其IC50 約為1046.7 μg/mL。在細胞凋亡方面,香蘭葉水萃取物處理的HepG2 促凋亡蛋白Bim、Bik 表現上升,Bax 沒有變化,抗凋亡蛋白Mcl-1 增加,凋亡相關的PARP-1 蛋白則沒有觀察到明顯的切割情形。本研究證實香蘭葉水萃取物對HepG2 有抑制細胞存活的效果,實驗結果可提供香蘭葉水萃取物做為肝癌臨床用藥可能性的參考。

Extract of leaves of Pandanus amaryllifolius performed anti-oxidative, anti-hyperglycemic, anti-viral, anti-neogenic, and apoptosis-promoting activities on different types of cells. In order to study whether bioactive compounds of P. amaryllifolius leaves exert therapeutic activities on liver cancer, we acquired water extract of P. amaryllifolius leaves by ultrasonic extraction and analyzed the effects of the extract on survival and apoptosis of hepatocellular carcinoma HepG2 cells. Our results showed that survival of HepG2 cells was inhibited by treatment of water extract of P. amaryllifolius leaves. After HepG2 cells were treated with the extract, pro-apoptotic Bim and Bik as well as anti-apototic Mcl-1 protein expression was increased. No change in Bax expression and PARP-1 cleavage was observed after the extract treatment. This study showed that the water extract of P. amaryllifolius leaves inhibits HepG2 cell survival and provides an evidence for further study of the extract as a clinical treatment option for hepatocellular carcinoma.
