摘要


自1990年7月至1991年6月,36個經開刀手術取出之尿路結石,分別經由紅外線光譜儀分析其晶體成份;以及運用單向和雙向電泳分析法(one and two-dimensional gel electrophoresis) 分析結石之蛋白質成份。結石晶體之成份分析顯示:其中單水草鈣5例,尿酸4例,磷酸銨鎂2例,磷灰石2例,單水草酸鈣+磷灰石3例,單水+雙水草酸鈣2例,磷酸銨鎂+磷灰石3例,單水+雙水草酸鈣15例。不同結石成份含有不同多寡之蛋白質含量,其中磷灰石含量最多,每克磷灰石成含45.61± 17.20微克(ug)蛋白質,尿酸居次24.94±16.57微克/每克結石,接著是磷酸銨鎂+磷灰石21.84±12.03,單水+雙水草酸鈣19.09±3.34,單水+雙水草酸鈣+磷灰石18.10±13.44,磷酸銨鎂16.50 ± 10.72,單水草酸鈣14.77 ±14.30。單向和雙向電泳分析法之結果是一致,其結果顯示尿路結石之蛋白質分子量自小於14 kDa 至 18 kDa。再者,不同成份之結石具有不同之蛋白質組成,此項研究結果暗示蛋白質在尿路結石形成之過程中可能扮演一積極且選擇性地角色而促使結石形成。總結而言,運用單向和雙向電泳分析法分析尿路結石蛋白質之成份是相當好的一種研究方法,不同晶體成份之尿路結石亦具有不同成份和多寡之蛋白質。

Between july 1990 and june 1991, 36 urinary calculi removed by open surgery were analyzed by in fraire spectrophotometer. The protein content of these stones were analyzed by one and two-dimensional gel electrophoresis. The stone composition of 36 urinary calculi were whewellite in 5, uric acid 4, struvite 2, carbonate apatite 2, whewellite + carbonate apatite 3, whewellite + weddllite 2, struvite + content in different urinary calculi was varied, carbonate apatite contained more protein than other composition: 45.61± 17.20 ug protein/g stone, followed bue uric acid 24.94±16.57, struvite+ carbonate apatite 21.84±12.03, whewellite + weddellite ±19.09 ±3.34, whewellite + weddellite + carbonate apatite 18.10±13.44, struvite 16.50 ± 10.72, whewellite 14.77 ±14.30. The result of the protein map analyzed by one and two-dimensional electrophoresis from the same stone was consisent, which showed a molecular weight pattern ranged from less than 14 kDa to 18 kDa. Furthermore, different urinary calculi had a distinct protein map. This finding strongly suggests that protein may play an active role in selective incorporation of stone formation. In conclusion, one and two-dimensional electrophoresis is a novel method to analyze urinary calculi protein, the results are characteristic and reproducible.
