Blimp-1 Overexpression in Early T-Stage Gingival Cancer with Neck Metastasis-A Case Report and PDTX Pilot Study

早期牙齦癌合併頸部淋巴結構轉移及Blimp-1之過度表現-病例報告及腫瘤異體移植之試驗研究

摘要


上皮一間質細胞轉化在口腔癌的病程進展,諸如頸部及遠端轉移等,扮演著重要的角色。而Blimp-1因子藉由影響上皮一間質細胞轉化,進而促進部分癌細胞的移動與轉移。和傳統的癌細胞培養相較之下,腫瘤異體移植更能夠忠實的呈現腫瘤的結構及其細胞異質性。本篇病例報告一牙齦癌病患,其癌症病程雖較早期,卻已出現多顆頸部淋巴結侵犯。在組織免疫化學染色之下,發現Blimp-1過度表現的情形。我們使用腫瘤異體移植的方式將此高度侵犯性的癌症培養在大鼠身上並保留下來,可供作後續研究及可能的針對Blimp-1因子的口腔癌標靶藥物之研發使用。

The epithelial-mesenchymal transition is a crucial process in the successful execution of numerous processes, such as the metastasis of oral cancer cells. Blimp-1 may play a critical role in migration through epithelial-mesenchymal transition in oral cancer cells. Compared with conventional cancer animal models, patient-derived tumor xenograft (PDTX) models may more faithfully recapitulate the tumor architecture and cellular heterogeneity observed in patient tumors. In this case, multiple prominent lymph node metastases were noted, although it was a T1-stage gingival cancer. Blimp-1 overexpression was observed in this tumor after an immunohistochemical stain examination. A PDTX model was used to maintain this aggressive cancer. We want to test the potential therapeutic target of Blimp-1 via our employed PDTX oral cancer models in future studies.
