幾丁聚醣/槲皮素混摻水膠之製備及應用

摘要


槲皮素是一種黃酮類天然的抗氧化劑;具有多種生物活性,如抗發炎、抗病毒、抗癌和抑菌等。本研究將幾丁聚醣與醋酸纖維素混摻製備成水膠,並在水膠內包覆不同比例槲皮素。幾丁聚醣/醋酸纖維素水膠在不同pH值的緩衝液測試其膨潤性質,實驗結果顯示幾丁聚醣/醋酸纖維素水膠隨著pH值增加其膨潤度減少。幾丁聚醣/醋酸纖維-槲皮素水膠其抗氧化化活性以DPPH自由基清除率、亞鐵離子螯合能力與總酚含量等分析方法探討。最後以L929小鼠纖維母細胞觀察其生物毒性,其結果顯示混摻水膠未具有細胞毒性。綜合以上結果,幾丁聚醣/醋酸纖維-槲皮素水膠做為傷口敷料,極具有發展潛力。

Quercetin, a flavonoid type natural antioxidant, exhibits various biological effects, such as anti-inflammatory, anticancer, antiviral and antibacterial activities. In this study, chitosan (CS)/cellulose acetate (CS) blended hydrogels with various content of quercetin were prepared. The swelling behaviour of the chitosan/cellulose acetate (CS/CA) hydrogels was examined in buffered solutions at various pHs. The equilibrium swelling ratios of the CS/CA hydrogels decreased with increasing pH. The antioxidant activities of quercetin-loaded CS/CA hydrogels were evluated by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH), ferrous iron chelating and total phenolic content assays. In addition, the quercetin-loaded CS/CA hydrogels were proven non-cytotoxic to L929 mouse fibroblasts. These characteristics indicated that the quercetin-loaded CS/CA hydrogel could be a promising canditdate for wound dressing.
