包覆虎杖萃取物之蠶絲蛋白/幾丁聚醣奈米纖維的製備與特性研究

摘要


根據文獻報導虎杖根具有多種藥理活性,包括抗氧化、抗發炎、抗癌、抗病毒、抗真菌和抗細菌等。本研究將蠶絲蛋白與幾丁聚醣混摻並包覆虎杖萃取物,利用靜電紡絲的方式製備蠶絲蛋白/幾丁聚醣-虎杖萃取物奈米纖維膜。此蠶絲蛋白/幾丁聚醣-虎杖萃取物奈米纖維膜經由掃瞄式電子顯微鏡觀察其表面型態、雷射掃描共軛焦顯微鏡測試其螢光性質、清除DPPH自由基試驗分析其抗氧化活性及MTT分析方法評估對L929小鼠纖維母細胞之細胞毒性。使用高效能液相層析儀(HPLC)測定此奈米纖維薄膜在37℃下醋酸緩衝液/乙醇之虎杖萃取物其活性多酚化合物釋放行為。實驗結果顯示,包覆虎杖萃取物之蠶絲蛋白/幾丁聚醣奈米纖維膜具有應用於傷口敷料之潛力。

The root of Polygonum cuspidatum has been reported to possess many pharmacological effects including anti-inflammatory, antioxidan, anticancer, antiviral, antibacterial, and antifungal effects. In this study, silk fibroin (SF)/chitosan (CS) composite nanofibrous mats containing Polygonum cuspidatum extract (PCE) were prepared via electrospinning process. The morphology and fluorescent properties of the SF/CS-PCE nanofibrous mats were characterized by scanning electron microscopy (SEM) and laser scanning confocal microscopy (LCSM). Antioxidant activity of SF/CS-PCE nanofibrous mats was measured by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Indirect cytotoxicity of SF/CS-PCE nanofibrous mats was evaluated using L929 fibroblasts by the MTT assays. The release behavior of bioactive polyphenols in PCE from composite nanofibrous mats was investigated in acetate buffer solution/ethanol 37℃ using high-performance liquid chromatography (HPLC). These results suggest that the PCE-loaded SF/CS nanofiber mats are promising for wound dressings.
