洛神花萼萃取物之酵母菌發酵物對小鼠B16F10黑色素瘤細胞生成黑色素之抑制

本研究探討乳酸克鲁維酵母菌(Kluyveromyces lactis),進行洛神花萼萃取物發酵是否具有加成效果。將發酵24、48及72 hrs後的酒精萃取物與水萃取物,分別加入B16F10黑色素瘤細胞內培養,測試其發酵時間對於抑制黑色素生成之效果,探討洛神花萼發酵萃取物是否具有美白的加成功效。結果顯示:洛神花萼發酵萃取物的發酵時間長短,對於抑制黑色素生成並無顯著差異。因此我們選擇發酵24 hrs之後的酒精萃取物,與其他植物進行比較,例如:紅豆、薏仁、山苦瓜對黑色素生成的抑制效果,結果以洛神花萼顯著勝出。當洛神花萼酒精萃取物濃度於100 μg/mL與200 μg/mL時,影響黑色素生成量分別為52 ± 4%與42 ± 7%,顯著低於發酵水萃取物(94 ± 4%;91 ± 7%)與未發酵者(85 ± 4%;78 ± 8%),顯示其抑制黑色素效果卓越。甚至比市售麴酸(80 ± 6%;73 ± 5%)的美白功效更加優越。總結:經過發酵的洛神花萼酒精萃取物,具有顯著抑制黑色素生成之功效,推測其具有應用於美容保健市場之潛力。

In this study, we selected Kluyveromyces lactis (K. lactis) to ferment with Roselle calyx for 24, 48 and 72 hrs. We supposed that the ferment extract could be additionally powerful to inhibit the melanin synthesis. Based on the results, there was no significant difference of fermentation time for the ferment ingredients to inhibit the melanin production. Moreover, we compared ethanol extracts of Roselle calyx, red bean, coix seed, and Momoridica dioica with fermentation for 24 hrs to examine the anti-melanogenic effect in B16F10 melanoma cells. It showed that Roselle calyx was the best inhibitory ability. Furthermore, the ethanol extract of Roselle calyx, at 100 and 200 μg/mL, with K. lactis fermentation for 24 hrs, has much lower melanin levels (52 ± 4% and 42 ± 7%) than that without fermentation (85 ± 4 %;78 ± 8 %), aqueous extract (94 ± 4 %;91 ± 7 %), and Kojic acid (80 ± 6 %;73 ± 5 %). In conclusion, we suggested that the ethanol extract of Roselle calyx ferment has good activity to inhibit melanin synthesis, which has potential to develop a novel whitening healthy product.
