以基因晶片分析中藥黃芩培養的腎細胞的基因表達

Analyzing Expressed Genes of HEK Cells Cultured with Scutellaria Baicalensis Georgi by Gene Chips

摘要


本研究是從基因晶片的層面來探討中藥黃芩與HEK 293(human embryonic kidney cell)腎細胞株一起培養,抽取mRNA,反轉錄為cDNA,再與白果能博士實驗室的晶片上的已知探子基因晶片進行雜合反應,並利用單色酵素呈色法呈色後(為藍色)放入彩色掃瞄器中進行掃瞄及影像資料分析以基因微陣列觀察基因的變化,並與未加中藥的腎細胞株做比較,試圖找出能影響HEK 293腎細胞相關的基因。 結果發現黃芩處理後的細胞基因表達,大於兩倍標準差的共有21個基因,小於負兩倍標凖差的共有386個基因。其中下調的基因包括Ubiquionl-cytochrome coreductase binding protein, cytochrome coxidase, subunit VIa, polypeptide, 1 calmodulin, major histocom-patibility complex, classI, B,而上調的基因包括erythropoietin receptor, parathyroid hormrne-like hormone。未來研究將進一步了解其意義之所在。

In this study, we try to elucidate the gene expression of HEK 293 cells treated with Chinese herbal powder produced from Scutellaria baicalensis Georgi. For this, gene expression of herbs treated cells and untreated cells were compared simultaneously using Dr. Peck Konan microarray system. Dual color staining method of enzyme colorimetry detection system was utilized by biotin-16-dUTP during reverse transcription. A color microarray image was rendered by the chromogens yielded by the enzymatic reactions. The result showed changes in expressions of 21 genes up regulation and 386 genes down regulation by Scutellaria baicalensis Georgi. The 386 genes down regulation include Ubiquinol-cytochrome C reductase binding protein, cytochrome C oxidase, subunit VIa, polypeptide, calmodulin, major histocompatibility complex, class I, B. The 21 genes up regulation include enythropoietin receptor and parathyrathyroid lormone-like hormone. This study established a method to the understanding of the genetic influence of TCM (Scutellaria baicalensis Georgi). Further studies are needed to verify these results.
