Effects of Cadmium on the Libido and Fertility of Male Mice

鎘對雄鼷鼠之性慾與授孕能力的影響

己知重金屬如鉛或鎘會引起雄性生殖系統的病變,然而是否這些有毒的作用對活的生殖細胞能造成任何遺傳的致害尚無充分研究。本研究旨在使用顯性致死試驗(Dominant lethal test)來分析並評估鎘對雄鼷鼠之生殖細胞的影響。雄鼷鼠經腹腔注射單一劑量每公斤體重5mg或2mg氯化鎘後分別在第二與第三週喪失性慾及授孕能力。注射氯化鎘後2至4天,可見嚴重的顯性致死效應。雄鼷鼠給予每公斤體重2mg氯化鎘後自六週起恢復性慾及授孕能力,雖可交配受孕但母鼠並無活胚存在。此外,授孕能力大約再過一週減退,這表示在此低劑量的鎘濃度下,雄鼷鼠的B型精原母細胞(Spermatogonia type B)對隔離子會造成耐性,但在此同時,顯性致死突變也會累積,並達殺死胚。

Heavy metals such as lead or cadmium are well known to cause lesions of the male reproductive system. Whether these deleterious effects can produce genetic hazards of surviving germ cells is poorly known. We made dominant lethal tests of the effect of cadmium on germ cells of male mice. Mice injected intraperitoneally with cadmium chloride (a single dose 5 mg/kg or 2 mg/kg) lost their libido and fertility in the second and third week respectively. Before this (2-4 days after administration), severe dominant lethal effects were noticed. Mice given cadmium chloride (2mg/kg) showed libido and fertility from the sixth week after exposure but no live embryos were born from mating. Moreover, the fertility declined after about a further week. This behaviour indicates that at this dosage mouse spermatogonium type B cells can become tolerant to the cadmium ion, but at the same time dominant lethal mutations also accumulate and kill the embryos.
