Liquid Chromatographic Determination of Sulfonamides in Feeds and Pork Integrated with Postcolumn Derivatization

摘要


本研究之目的在嘗試開發出一個簡單和快速檢測飼料和豬肉中磺胺劑之方法。利用高效法相層析法搭配後量反應,將磺胺劑與P-dimethylaminobenzaldehyde反應後於波長450 nm進行檢測。結果顯示飼料及豬肉樣品僅須各別經甲醇鹽酸溶液及氰甲烷溶液處理後,即可進行分析,利用Intersil ODS-2分離管,以30%氰甲烷溶液加0.05%三氟醋酸緩衝液為移動相,流速0.5 mL/min下進行分析,其藥物滯留時間分佈在6至16分鐘之間,且無干擾物質,故可成功地自飼料及肉中檢測出磺胺劑。飼料添加磺胺劑時之回收率介於90.2-94.1%之間;豬肉添加回收率亦達86.3-107.3%。兩者最低檢測界限為0.01 ppm。

A simple, rapid, and reliable method for determination of sulfonamides in feeds and pork was developed. Samples were extracted by 0.1 N metholic acid and acetonitrile from feeds and pork, respectively. Sulfonamides were chromatographice on a Intersil ODS-2 column (4.6 x150 mm) using acetonitrile-0.05% trifluroacetic acid (30: 70) as the mobile phase. The sulfonamides were detected at 450nm after postcolumn reaction with P-dimethylaminobenzaldehyde, and their retention times ranged from 6 to 16 min without any interference. The recoveries of spiked samples were between 90.2-94.1% in feeds and 86.3-107.3% in porks, respectively. The detection limits of residual drugs were 0.01 μg/g.
