中藥「乙肝康」對豬傳染性胃腸炎病毒在細胞培養時感染力的影響

Effects of a Chinese Herbal Medicine Yi-Kang-Kong on TGEV Infectivity in Tissue Culture

摘要


中藥「乙肝康」為台灣本土所研發之新型方劑,為數種草藥翠取物以不同比例混合而成。為了解該中藥在獸醫學可能的應用,我們用傳染性胃腸炎病毒(TGEV)的豬睪丸細胞組織培養系統為研究模式,探討該中棄對TGEV感染力的影響。籍由TCID50的測量發現中藥與病毒混合後,在不同濃度中藥與作用溫度,病毒對細胞的感染力均受到相當程度之抑制。由濃度與細胞數的關係,得到中藥對細胞之ID50為28.8µg/mL。TGEV經中藥低於ID50的三個濃度(6.3,10及25用µg/mL)混合作用,其TCID50分別為對照組之12.07±2.85%,3.02±0.78%和8.84±1.07%;顯示中藥濃度與病毒感染力之抑制,並非呈線性關係。若中藥與病毒分別在4,25,37℃作用,發現只有在25與37℃時病毒感染力受到抑制,其TCID50分別為對照組之44.53±9.24%,3.02±0.42%;顯示在生理溫度時中藥的抑制作用最高。考慮中藥對細胞前處理對病毒感染力的影響,發現細胞分別經中藥2或8小時處理後,病毒感染力受到相同程度抑制,其TCID50約為對照組之四分之一。這點表示中藥的處理可改變細胞性質使病毒對細胞的感染受到抑制,此過程在處理2小時內即可完成。

A Chinese herbal medicine, yi-kang-kong, composed of several herbal extracts was recently developed in Taiwan. To test its possible application in veterinary medicine, we used the TGEV/ST cell model to investigate the effect against viral infections of yi-kang-kong. By measuring the TCID50 as an infectivity index, we found yi-kang-kong could decrease viral infectivity, depending on the concentration of yi-kang-kong and the incubation temperature with the cells. Yi-kang-kong is toxic to cultured ST cell line at ID50 of 28.8µg/mL. The infectivity of TGEV decreased 12.07±2.85%, 3.02±0.78%, and 8.84±1.07% compared with that of the control group when mixed with yi-kang-kong in concentrations of 6.3µg/mL, 10µg/mL, and 25µg/mL, respectively. The infectivity of TGEV decreased at 25℃ and 37℃ by 44.53±9.24% and 3.02±0.42%, respectively, in the presence of 10μg/mL of yi-kang-kong. This result indicates that yi-kang-kong could best inhibit TGEV infectivity at normal physiological temperature. When cells were pretreated with yi-kang-kong for 2 or 8 hours, the infectivity of TGEV could be decreased by about 25%. This result indicates that yi-kang-kong pretreatment can strengthen cells against TGEV infection.
