橘色萬代蝶蘭微體繁殖之研究

Studys on in-vitro propagation of orange Vandaenopsis cultivars

摘要


蝴蝶蘭近年來利用異屬雜交技術創造出許多極具發展潛力之新品種,然異屬雜交蝴蝶蘭類遺傳生育特殊,難以大量繁殖生產,本研究利用不同培植體及生長調節劑進行瓶內增殖,建立微體繁殖技術。萬代蝶蘭Vandaenopsis Irene Dobkin 'Elmhurst'與Vandaenopsis Pulcherrimin莖頂培養於添加TDZ及BA組合NAA的1/2 MS培養基中皆可增殖,又以1、2 mg.L^(-1) TDZ處理有較高之存活率、擬原球莖體(protocorm like body, PLB)發生率及總增殖率。葉片切段培養後,則只有帶葉鞘的葉片基部有PLB形成,葉片培養前先浸泡硝酸銀溶液則可提高葉基培植體的增殖倍率。繼代培養基中添加1 ~ 2 mg.L^(-1) zeatin,或香蕉粉等有機添加物有助於PLB及芽體繼續增殖發育。現有技術:蝴蝶蘭及萬代蘭組織培養技術。創新內容:成功建立異屬雜交萬代蝶蘭品種之擬原球莖體誘導方式。對產業影響:可用於萬代蝶蘭新品種量產。

In the recently years, more and more Phalaenopsis new cultivars were developed by using intergeneric hybrid techniques. These hybrids are normally hard to propagate massively due to genetic problems and growth habits. This study aimed to establish primary in vitro propagation technique by using different explants and plant growth regulators for intergeneric Phalaenopsis hybrids multiplication induction. Shoot tips of Vandaenopsis Irene Dobkin 'Elmhurst' and Vandaenopsis Pulcherrimin could regenerate in 1/2 MS medium incorporated with TDZ or BA combined NAA treatments. Higher survival rate, PLB (protocorm-like-body) induction rate, and total proliferation were observed in 1 or 2 mg.L^(-1) TDZ treatments. Different leaf segment explants were cultured and only the basal part with leaf sheath had PLBs formation after culturing. Soaking leafs in silver nitrate solution before cultured could increase proliferation of leaf base explants. Adding 1-2 mg.L^(-1) zeatin, or organic complex like banana powder in sub-cultural medium could promote PLBs and shoots to proliferate and develop. What is already known on this subject? In vitro propagation techniques of Phalaenopsis and Vanda. What are the new findings? Establish a protorm-like-body induction process in intergeneric hybrid cultivars of Vadaenopsis. What is the expected impact on this field? This study can use in mas propagation of Vadaenopsis new cultivars.
