小家鼠(Mus musculus)標本毛色與標本製作法的關聯性分析

An Analysis of Coat Color of Museum Skin Specimen of Mus musculus in Relation to Preparation Methodology

摘要


我們使用Photolmpact繪圖軟體分析485隻於臺灣探集之小家鼠(Musmusculus castaneus)之毛色變異,分別用4個毛色測量值(明度、紅色、綠色、藍色色頻)代表背部與腹部的毛色,接著利用主成份分析法分析15個採集地捕捉的小家鼠,得到2個主成份(分別代表小家鼠毛色的明暗以及背腹毛色差異)以進行毛色分析。我們發現小家鼠背部與腹部的典型毛色皆為褐色,但腹部毛色較背部淡。小家鼠毛色在不同性別、採集年份、採集季節、採集地點與體型之間沒有不同,但標本製作方法會影響標本的毛色:攤開式標本的毛色較亮,冰棒狀標本的毛色較暗。我們認為這是因為攤開式標本在製作與保存過程中,缺乏有效的脫油程序, 故出油情況較嚴重所造成的結果。本研究建議未來學者利用標本進行毛色研究時,需考量標本製作方法對毛色的可能影響。

Coat color variation of 485 house mice (Mus musculus castaneus) in Taiwan was measured using Photolmpact graphic software. Four properties of coat color (luminosity, red, green and blue color channels) were measured on dorsal and ventral skin specimen. Two variables, represent lightness and dorsal-ventral color contrast, respectively, were derived from a principal components analysis from 15 localities. Both dorsal and ventral typical coat colors are agouti, but ventrum was generally lighter than dorsum. The coat colors are not significantly different between sexes, years, seasons, localities, and body sizes. However, the methods of making specimen significantly influence the coat color. The flat skins are lighter than stuffed study skins. We suggest that because fiat skins leaked more grease than stuffed study skin, which has cotton or other materials to absorb the grease. The results indicate that extra caution must be taken to remove as much grease as possible to minimize the bias in measuring color variation.
