TGF-ß1迷你豬牙周組織實驗性炎症中之含量變化

TGF-ß1in the Experimentally Induced Inflammatory Periodontal Tissues in Miniature Swines

摘要


Transforming growthe factor-ß1(TGF-ß1)是多重功能之分子,它對許目標結胞及組織且有獨特極強烈的作用;可經由某此細胞間的互相作用而促進炎性反應;另外,在極微量下即具有極強的單核吞噬細胞趨作用,亦可因增加PGE2之產生而促進骨吸收。另一方面,TGF-ß1可能藉著調整其他生長因子對matrix metallopeinases(MMPs)及tissue inhibitor of metalloproteinases (TIMPs)表現的機轉來擔任細胞間質更換之極重要角色。已有研究證實在發炎的牙齦組織細胞內外皆有TGF-ß1之存在,其分佈也只限於有炎性反應的區域。本研究是利用16隻迷你豬的牙齒做牙炎/牙周炎之誘導,實驗組於第一至第三天連續作Porphyromonas gingivalis及Actinomyces viscosus 19246的口腔內感染。於第三、五、八、十三週特定時間內比較兩組個項臨床指數及牙齦溝內結紮絲線上之黑色菌落比例之變化,並以酵素免疫分析法測定實驗組及對照組牙齦組織中TGF-ß1之含量,期能了解這種介質含量變化與疾病狀態之關系。結果顯示實驗組TGF-ß1濃度均大於對照組,且在第三週時,二者有統計學上的差異;第三週和第十三週間實驗組TGF-ß1之濃度有顯著差異且與結紮時間呈負相關性。實組TGF-ß1之濃度與臨床指數-牙結石指數,牙齦指數變化量呈負相關性。綜合以上結果顯示TGF-ß1之濃度隨著實驗性牙齦組織炎症之狀況而有動態變化,因此建議可利用牙齦組織中TGF-ß1含量變化來探討牙齦組織發炎情形。

TGF-ß1 is a multifunctional molecule which has unique and potent effects on many target cells and tissues. TGF-ß1 may promote in flammatory reaction by certain interecellular interaction. TGF-ß1at extremely low concentrations shows strong chemotatic activity for mononuclear phagocytes and stimulates bone resorption by enchancing production of PGE2. On the other hand , TGF-ß1 plays a very important role in the regulaton of modulating the action of other growthe factors on matrix metalloproteinases(MMPs) and tissue in hibitor of metalloproteinases (TIMPs) expression. TGF-ß1 was identified intra- and extracellularly in the infflamed gingival tissues and the distribution was associated with areas of inflammation. Sixteen miniature swines were used in this experimental gingivitis/perodontitis study. The ligatures were placed in situ for preiods of 3,5,8 and 13 weeks and peroral innoculations of Porphyromonas gingivalis| Actionomyces viscosus into the ligatures were carried out only in the expermental group.ELISA was used to measure the levels of TGF-ß1 in gingival tissues from the experimental and control groups. Recording of the clinical periodontal parameters was performed and the proportion of black-pigmented Bacteroides in the ligature(plaques) removed immediately prior to the biopsies was recorded. The results revealed that the concentration of TGF-ß1 of the experimental group was higher and significantly different in statistics on the period of third week than that of the control group. The concentration of TGF-ß1 was significanty different between the third week and the thirteenth week tin the experimental group, and was negatively realted to the time-length of ligatures. Furthermore, the concertration of TGF-ß1 was negatively related to the changes of the calculus index and gingival index. These data indicated that the concentration of TGF-ß1of gingival tissue exhibited dynamic changes associated with the progression of experimental periodontal inflammation. The levels of TGF-ß1 in gingival tissue may be valuable in detecting the inflammatory reaction of periodontal tissues.
