Medium Effects on the Increase of Protease and Cellulase Activities of Bacillus subtilis BCRC14716

培養基組成對Bacillus subtilis BCRC14716蛋白酶與纖維素酶活性之影響

摘要


以TSB培養基為基礎,額外添加CMC、葡萄醣胺或蝦殼粉之複合培養基(skim milk 3.4%, soybean meal 0.6%, glucose 0.5%, NaCl 1%, K2HPO4 0.5% and MgCl2 0.5%)誘導Bacillus subtilis BCRC14716生產高活性蛋白酶及酶纖維素酶,該酵素液與基質反應溫度以50℃較37℃佳。該菌株以另添加葡萄醣胺組分別培養48與72小時後,得到高活性蛋白酶與酶纖維素酶,酶其活性分別為47 U/mL及222 U/mL。溶於1%醋酸緩衝溶液之幾丁聚醣與酵素液混合,於37℃或50℃水解1小時,以HPLC測定水解產物寡醣總量,則以50℃處理組可得最高含量幾丁寡醣(70713 ppm)。

A modified medium containing skim milk 3.4%, soybean meal 0.6%, glucose 0.5%, NaCl 1%, K2HPO4 0.5% and MgCl2 0.5% was prepared based on tryptone soya broth (TSB) medium. The carboxyl-methyl cellulose (CMC), glucosamine or shrimp shell powder were added to induce the cellulase and proteases of Bacillus subtilis BCRC 14716. Enzyme activities with reaction temperature at 50℃ were higher than those at 37℃. The highest activities of protease (47units/mL) and cellulase (222units/mL) were observed in those cultured in the modified medium with glucosamine for 48 and 72 h, respectively. When the chitosan in 1% acetate buffer (pH 7.3) was hydrolyzed by the enzymes obtained from 3-days cultured Bacillus subtilis BCRC 14716 at 37º and 50℃ for 60 min, the total yield of chitooligosaccharides was 70713 ppm after 60 min hydrolysis at 50℃.
