Occurrence of Tetrodotoxin in Two Gastropods from Philippines by Using HPLC and LC-MS/MS Methods

摘要


河魨毒(TTX)為常見的海洋生物毒,其毒作用機制為阻塞神經細胞上之鈉腔體,導致神經訊息無法傳遞。西元2005年在菲律賓宿霧島沿岸採集兩種腹足綱螺類百肋鳳凰螺及大法螺,經老鼠生物毒性檢測法顯示河魨毒存在於螺類之中腸腺。兩種螺類分別平均毒性百肋鳳凰螺為28±19(mean±S.D.)及大法螺為52±17MU。所有樣品毒素之萃取經C18固相萃取法及3,000道爾頓分子篩過濾。經高效液相層析法(HPLC)及液相層析串聯式質譜儀法(LC-MS/MS)偵測。證明毒素主要為河魨毒,麻痺性貝毒(PSP)並未被檢測出。河魨毒在菲律賓海域之螺貝中,其食用安全性值得注意。

Tetrodotoxin (TTX) is widely known marine neurotoxin, which acts by blocking the entry of sodium ion into sodium channels of neuronal membranes. Toxicity was determined in two gastropods (Strombus labiatus and Charonia tritonis) from Cebu Island, Philippines on October 2005, showing lethal potency in the digestive gland of gastropod by using TTX bioassay. The average toxicity of gastropod specimens was 28±19 (mean ± S.D.) and 52 ± 17 MU/specimen in S. labiatus and C. tritonis, respectively. All toxic extracts of each species samples were purified by passing through C18 Sep-Pack solid phase extraction column and filtered through a microcentrifuge filter. The eluate was filtered through a microcentrifuge filter. The filtrate was freeze-dried, dissolved in distilled water, and determined by high performance liquid chromatography (HPLC) and liquid chromatograph tandem mass spectrometry (LC-MS/MS). The results demonstrated that the toxin contained TTX. The paralytic shellfish poison (PSP) was not detected. The evidence of TTX in both gastropods warns of potential problems in the Philippines gastropods.
