金花石蒜之鱗片組織培養繁殖

Tissue Culture Propagation of Bulb-Scale of Lycoris aurea Herb.

摘要


於不同秀節行金化石蒜魚片培養(bulb-scale culture),以5月反採之培植體出芽率最高;8~9月出芽率最低,且較其他秀節褐化率高。鱗片培養以MS爲基本培養基加BA 2~3 mg/ℓ,NAA 0.5 mg/ℓ爲促進出芽最適之培養基。在27±2℃,日光燈2000 lux下培養,2個月後不定芽形成,之後將繼代培養次減少中移至無任何生長素的培養基,在20±1℃,1600 lux照光培養,有利小鱗莖形成及發根。 由一個鱗莖(大小約8×5 cm)之魚片培養經7~8個月,平均可得10 cm高之50~75小植株;若由鱗片培養所得不定芽去其頂(decapitation)或在5月出芽高之季節行培養,將可得100植株或更多。

Bulb scales of Lycoris aurea Herb, were cultured year-round in ordor to find the best time for excision, Scales cultured in May gave the highest percentage of explants producing adventitious buds and these cultured in August and September the least, and the highest percentage of browning of scale explants. Murashige and Skoog medium (1962) supplemented with 2 or 3 mg/l BA and 0.5 mg/l NAA gave the highest percentage of survival under day-light fluorescent tubes with light intensely of 2000 lux under a light-dark cycles of 16/8 hours at 27±2℃. Adventitious buds appeared in the axile of the twin-scale explant after 2 months of culture. Transferring the scale explant with adventitious buds to medium without any growth regulator and incubation at 20±1℃ promoted the development of adventitious buds into plantlets with a bulblet due to the thickening of leaf base. From the twin scale explants excised from a mature bulb of 5 cm diameter and 8 cm height, 50-75 plantlets could be regenerated. Regeneration of more than 100 plantlets from a mature bulb is possible provide the excision is done in May and decapitation of regenerated bulblets was used to hasten the formation of adventitious buds.
