以秀麗隱桿線蟲為模式生物探討相思樹嫩枝萃取物之生物體內抗氧化

摘要


相思樹(Acacia confusa),又名臺灣相思,為豆類含羞草亞科金合歡屬植物,原產於臺灣南部,在日治時期被廣泛種植於臺灣。本研究利用模式生物秀麗隱桿線蟲(Caenorhabditis elegans)探討相思樹嫩枝萃取物對於生物體內抗氧化作用。生物體外的DPPH自由基清除能力試驗顯示,相思樹嫩枝萃取物具有明顯的自由基清除能力。生物體內的研究結果顯示,在氧化壓力誘導劑核桃酮(Juglone)誘導的氧化壓力下,相思樹嫩枝萃取物可以顯著提升線蟲的存活。本研究結果更進一步顯示,相思樹嫩枝萃取物可能經由降低生物體內的自由基/活性氧(Reactive oxygen species,ROS),而提高線蟲對於氧化壓力之抗性與適性。

Acacia confuse, also named Formosa Acacia (Taiwan Acacia), belongs to Fabaceae family and Acacia genus is a perennial tree native to South Taiwan. In the present study, A. confusa twigs extract was evaluated for its free-radical scavenging properties using both in vitro and in vivo assays. In vitro DPPH assay showed that A. confusa twigs extract was capable of scavenging free radicals. Further in vivo assay in Caenorhabditis elegans showed that A. confusa twigs extract (10 μg/mL)-treated worms exhibited an increased survival rate under juglone-induced oxidative stress and lowered intracellular reactive oxygen species level compared to that of the untreated control. Our study suggests that the protective effects of A. confusa twigs extract were likely due to its free radical scavenging ability thereby enhancing oxidative stress resistance.
