鳳梨癒合組織體胚形成過程中離層酸之分佈

Abscisic Acid Distribution during Somatic Embryogenesis in Pineapple Callus

摘要


鳳梨(Ananas comosus L. Merrill)是世界上重要熱帶果樹之一,本研究利用已建立之台灣食用鳳梨癒合組織植株再生系統,探討體胚形成與植株再生過程中離層酸(abscisic acid; ABA)之分佈及其可能扮演的角色。試驗選取台農20號(牛奶鳳梨;TN-20)之腋芽培養於同時添加NAA和BA之MS基礎培養基中,分別於不同培養天數取樣觀察形態變化,並進行ABA免疫組織化學定位分析。結果顯示培養後約5至6週可順利誘導癒合組織形成,繼代培養後2週可誘發體胚形成,約4週後可觀察到芽體形成。此外,在培養2週的組織中在導管系、原始分生帶、擬分生組織及新形成的體胚組織中呈現明顯ABA免疫反應,隨著體胚發育成芽體,也可在小芽附近觀察到ABA呈色反應。上述結果顯示ABA的分佈與鳳梨細胞分化可能有密切相關。本研究是第一篇利用免疫組織定位分析技術探討ABA與鳳梨體胚形成有關之報導,雖然ABA在這方面扮演的角色為何仍不清楚,須進一步試驗印證之,不過推測ABA可能誘發與體胚形成或芽體分化有關的基因表現,甚值得進一步深入探討。

To determine the relationship between abscisic acid (ABA) and somatic embryogenesis in pineapple (Ananas comosus L. Merrill cv. Tainung 20) callus, the distribution of ABA in pineapple during callus formation and shoot regeneration was studied by immunohistochemical analysis. The results showed that callus could be induced after five to six weeks when the axillary bud inoculated on MS basal medium containing NAA and BA. The somatic embryo and shoots were further regenerated after subculture for two weeks and four weeks, respectively. At the same time, we found the distribution of ABA was closely related to regenerative tissues, ex. tracheary elements, meristematic belt, meristemoid, and globular structure of somatic embryo. In our knowledge, this is the first paper to discuss the relationships between ABA distribution and early somatic embryogenesis in pineapple. The possible roles of ABA during induction of somatic embryogenesis and shoot regeneration in pineapple are further discussed.
